摘  要:目的建立酸枣仁汤及提取物中多糖含量的HPLC-ELSD测定方法并分析酸枣仁汤中多糖的含量。方法采用HPLC-ELSD法。以葡萄糖为对照品,测定酸枣仁汤总多糖及提取物中葡萄糖的含量,求出换算因子,通过计算样品中葡萄糖的含量换算出多糖的含量。测定条件:色谱柱为KromasilC18柱(250mm×4.6mm,5μm),流动相为甲醇-水(体积比5:95),流速为0.8mL·min^-1,ELSD漂移管温度为115℃,气体流速为3.1L·min^-1,雾化温度为113.4℃。结果酸枣仁汤中总多糖样品中多糖含量质量分数为20.23%,分子质量小于10ku的多糖样品SDP-1中多糖含量质量分数为11.80%,分子质量为10-50ku的多糖样品SDP-2中多糖含量质量分数为9.74%,分子质量大于50ku的多糖样品SDP-3中多糖含量质量分数为13.80%。结论建立的HPLC-ELSD测定方法快速、灵敏、操作简便、结果可靠,可作为酸枣仁汤及其提取物中多糖的含量测定方法,为其质量控制提供参考依据。Objective To develop an HPLC-ELSD method for the determination of the contents of polysac- charide in Suanzaoren decoction and extracts. Methods Polysaccharide was assayed by HPLC-ELSD. The glucose was used as standard control, and the purified polysaccharide was hydrolyzed to the glucose for determining polysaccharide conversion factor. Through the conversation factor, the content of glucose was converted to the content of polysaccharide in different samples. The chromatographic separation was performed on a Kromasil C18(250 mm ×4. 6 mm,5μm) column with a mobile phase of methanol and water ( V: V= 5:95) at a flow rate of 0. 8 mL·min^-1 ;the ELSD tube temperature was set at 115 ℃ ;the gas flow rate was 3.1 L· min^ - 1 ; nebulizer was set at 113.4℃. Results The polysaccharide contents of TSDP, SDP- 1, SDP- 2 and SDP-3 were 20. 23 %, 11.80% ,9.74% and 13.80%, respectively. Conclusions The established method is sensitive, convenient and reliable. It can be used for the determination of the content of polysaccharide in Suanzaoren decotion and extracts, and provide a reference for the quality control of Suanzaoren decotion polysaccharide.
