摘  要:目的:研究系统性红斑狼疮(SLE)模型小鼠(MRL/lpr)经解毒祛瘀滋阴方治疗后的代谢组学差异,以探讨解毒祛瘀滋阴方治疗SLE的作用机制。方法:收集醋酸泼尼松治疗组、解毒祛瘀滋阴方治疗组、SLE模型组和昆明种C57BL/6J对照组小鼠的尿液样本,应用高效液相色谱-四级杆-飞行时间质谱联用系统(HPLC-QTOF/MS),对尿液样本进行代谢图谱分析。应用Mass Hunter和MPP软件处理数据,数据模式识别采用有监督的偏最小二乘法-判别分析(PLS-DA)。结果:代谢组学分析发现SLE模型小鼠存在较明显的氨基酸代谢紊乱,从尿液样本中鉴别出9种有重要影响的代谢标志物。醋酸泼尼松可以部分修复SLE模型小鼠失衡的苯丙氨酸代谢通路,而解毒祛瘀滋阴方能多维修复失衡的甘氨酸、苯丙氨酸和色氨酸代谢通路。结论:解毒祛瘀滋阴方通过多维修复失衡的氨基酸代谢通路对SLE模型小鼠发挥治疗作用。Objective: To investigate the mechanism of Jiedu Quyu Ziyin Recipe for treatment of the systemic lupus erythematosus (SLE) by studying the effect of Jiedu Quyu Ziyin Recipe on the whole metabolism of SLE mice. Methods: A high performance liquid chromatography coupled with Q-TOF/MS (HPLC-Q-TOF/MS) was employed to analyze metabolic characteristic of the urine samples of the prednisone acetate-treated mice, Jiedu Quyu Ziyin Recipe-treated mice, SLE mice (MRL/lpr) and control group mice (C57BL/6J). Software of Mass Hunter and Mass Profiler Professional (MPP) were used to process the data. A supervised mode of partial least squares-discriminant analysis (PLS-DA) was applied to recognize the data pattern. Results: There were obvious disorders of amino acid metabolism pathway in SLE model by using the PLS-DA. Nine compounds which had a significant contributing to classification were identified as biomarkers. Prednisone acetate could partly repair the unbalanced phenylalanine metabolic pathways of SLE mice. Jiedu Quyu Ziyin Recipe could repair the unbalanced metabolic pathways of glycine, phenylalanine and tryptophan from several dimensions. Conclusions: The Jiedu Quyu Ziyin Recipe had a good therapeutic effect on the SLE model mice by regulating the dysfunctions of amino acid metabolism pathway.
