摘 要:目的观察肾移植患者移植后血清中的色氨酸(Tyr)、酪氨酸(Trp)及苯丙氨酸(Phe)含量变化情况,并探讨其与肾功能损伤的关联。方法用高效液相色谱-荧光法(HPLC-FD)测定肾移植患者和健康对照组血清Tyr、Trp和Phe含量,用自动生化分析仪测定血清肌酐、尿素氮、尿酸、胱抑素C、β2微球蛋白含量。分析氨基酸代谢与肾功能之间的关系。结果肾移植后患者血清Tyr、Trp含量与健康对照组比较差异无统计学意义,分别为(63.93±13.75)vs(58.13±16.50)μmol/L和(35.27±7.82)vs(37.15±9.37)μmol/L,P>0.05。Phe含量高于健康对照组[(60.75±13.55)vs(38.19±10.85)μmol/L,P<0.01],其中CKD3期患者Phe浓度明显高于CKD1与CKD2期患者(P<0.01)。经相关分析发现,Phe浓度与胱抑素C、β2微球蛋白浓度存在相关性。结论肾移植后患者体内存在芳香族氨基酸AAA代谢变化,这种变化与肾脏功能的状态密切相关。Objective To observe serum tryptophan (Tyr),tyrosine (Trp) and pbenylalanine (Phe) concentration change in post-renal transplantation patients,and explore the correlation of aromatic amino acid metabolic change with renal function. Methods Kidney transplant patients and healthy people serum Tyr,Trp and Phe content were determined by high performance liquid chro- matography-fluorescence spectrometry (HPLC-FD),and automatic biochemical analyzer was used to detected creatinine, urea nitro gen,uric acid,cystatin C and β2-microglobulin content. The relationship between aromatic amino acid metabolism and renal function were analyzed. Results Compared with the healthy control group,the concentrations of serum Tyr and Trp in post-renal plantation patients had no significant difference [(63.93±13.75) vs (58.13±16.50) μmol/L;(35. 27±7. 82) vs (37.15±9.37)μmol/L,P〉 0.05]. The concentration of serum Phe significantly increased than the healthy control group [(60.75 ±13.55) vs (38.19 ±10.85) μmol/L,P〈0.01],which CKD3 period,patients Phe concentration was significantly higher than CKD1 and CKD2 period patients (P〈0.01). The correlation analysis showed that Phe concentrations correlated with cystatin C and β2-microglobulin concentration. Conclusion Aormatic amino acid(AAA) metabolic change can be observed in post-renal transplantation patients,which is closely related to the state of renal function.
