摘 要:目的:研究锰对大鼠体内生殖激素和乳酸脱氢酶(LDH)水平的影响,探讨锰对大鼠生精细胞caspase-3表达与生殖激素和LDH水平之间的关系。方法:雄性SD大鼠随机分为空白对照组,15mg/kgMnCl2和30mg/kMnCl。组。免疫组织化学检测睾丸caspase-3表达,放射免疫测定血清睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)含量,化学比色测定血清和睾丸LDH活性。结果:与空白对照组比较,各染锰组caspase-3阳性细胞率均升高,血清睾酮含量降低,FSH和LH含量升高,6周30mg/kg组血清LDH活性升高,各染锰组睾丸LDH活性降低。染锰剂量相同,6周与4周比较,caspase-3阳性细胞率和血清FSH、LH含量升高,血清和睾丸LDH活性分别升高和降低。染锰时间相同,高剂量组与低剂量组比较,caspase-3阳性细胞率和血清FSH、LH含量均升高,血清和睾丸LDH活性分别升高和降低。结论:染锰15mg/kg4周即可诱发大鼠血清睾酮含量降低,FSH和LH含量升高及睾丸LDH活性降低和血清LDH活性升高,可能是促进生精细胞caspase-3表达的重要原因。Objective: To study the expression of caspase-3 in spermatogenic cells and the contents of reproductive hormones, lactate dehydrogenase (LDH) of rats exposed to manganese and to discuss the relationship between them. Methods: Male rats were randomly divided into blank control, 15 mg/kg and 30 mg/kg MnC12 groups. The expressions of caspase-3 in testis were investigated by immunohistochemical methods, the contents of testosterone (T), follicle stimulating hormone (FSH) and luteinizing hormone (LH) in serum were measured by radioimmunoassay, and the activity of LDH in serum and testis were measured by colorimetrie analysis. Results: Compared with blank control groups, the caspase-3 positive cell rate was increased, and the contents of T were decreased, while FSH and LH were increased in every manganese exposure groups. The activity of LDH was increased in serum in 6 weeks 15 mg/kg group, and decreased in testis in every Mnc12 group. Among manganese exposure groups of same dose, the caspase-3 positive cell rate and the contents of FSH and LH in serum were increased. The activity of LDH was increased in serum but significantly decreased in testis in 6 weeks MnC12 groups than that in 4 weeks groups. Among manganese exposure groups of same time, the caspase-3 positive cell rate and the contents of FSH and LH in serum were increased, and the activity of LDH was increased in serum but decreased in testis in 30 mg/kg manganese exposure groups than those in 15 mg/kg groups. Conclusion: MnClz at a dose of 15 mg/kg could decrease the contents of T, increase the contents of FSH and LH in serum, and decrease the activity of LDH in testis and increase that in serum, which might be important causes of accelerating the expression of caspase-3 in rat spermatogenic cells.
