摘 要:目的通过比较栀子苷、人参皂苷Rg1单独或配伍使用对氧糖剥夺损伤的小鼠小胶质细胞炎症因子分泌平衡的调节作用的差异,明确药物配伍调节缺血损伤小胶质细胞稳态的保护作用。方法利用氧糖剥夺方法复制小鼠小胶质细胞拟缺血损伤模型,栀子苷,人参皂苷Rg1单独使用及配伍使用(通络救脑注射液)分别作用不同时间后,采用Griess Reagent法测定一氧化氮(NO)含量,采用细胞计数试剂盒检测细胞活性,采用免疫酶联法及蛋白印迹法检测TNF-α及TGF-β的含量及表达水平。结果配伍使用可在不影响细胞生存活性情况下显著减少NO释放,显著降低缺血损伤小胶质细胞TNF-α水平,升高TGF-β水平,两个有效成分发挥疗效的侧重点不同,配伍使用明显优于两个有效组分单独使用。结论栀子苷,人参皂苷Rg1单独或配伍使用对拟缺血损伤人小鼠小胶质细胞分泌炎症因子具有平衡的调节作用,其机制与升高TGF-β及降低TNF-α表达有关。Objective To clarify the protective roles of compatibility of geniposide and ginsen- oside(Rgl ) in regulating ischemia injured microglia homeostasis by comparing the difference in regulato- ry roles of geniposide, Rgl, or ginsenoside + Rgl in balancing secretion of oxygen glucose deprivation in- duced microglia inflammatory cytokines. Methods The mimic ischemia injured microglia model was in- duced by oxygen-glucose deprivation (OGD). Then geniposide, Rgl, or ginsenoside + Rgl (Tongluo Jiun- ao Injection, TJI) was respectively added. The NO content was determined by Griess Reagent. The cyto- activity was detected using cell count kit. Contents of TNF-α and TGF-β and their expression levels were detected by ELISA and Western blot. Results Geniposide + Rgl could significantly inhibit the release of NO, elevate the TGF-β level, and decrease the content of TNF-α without influencing the cell survival. The two active ingredients played different therapeutic roles. The compatible use was obviously superior to use any one of the two active ingredients alone. Conclusions Geniposide, Rgl, or Ginsenoside + Rgl had regulating roles in balancing ischemia injured microglia homeostasis. Its mechanisms might be relat- ed to up-regulating the TGF-β expression and down-regulating TNF-α expression.
