摘  要:目的探讨重度子痫前期患者胎盘Axl的表达及与其发病的关系。方法选择2012年4月至2012年10月在四川大学华西第二医院就诊的孕产妇。分别采用免疫组织化学法、蛋白印迹、酶联免疫吸附及实时荧光定量RT-PCR法检测胎盘Axl蛋白及mRNA的表达。采用方差分析、t检验等方法比较各组间胎盘Axl蛋白及mRNA的表达强度,采用Pearson或Spearman's法进行相关性分析。结果 (1)重度子痫前期患者及正常妊娠者胎盘母面及子面同时存在Axl蛋白的表达部位一致,均定位于胎盘的血管内皮细胞。(2)重度子痫前期患者及正常妊娠者各自胎盘母面及子面Axl蛋白的表达无统计学差异。(3)重度子痫前期患者胎盘Axl蛋白的表达水平显著高于正常妊娠组,差异有统计学意义(P<0.05)。(4)重度子痫前期患者胎盘Axl-mRNA表达水平显著低于正常妊娠者,差异有统计学意义(P<0.05)。(5)重度子痫前期患者胎盘Axl蛋白表达水平与分娩前收缩压、舒张压及24 h尿蛋白呈正相关,与分娩孕周呈负相关;正常妊娠者胎盘Axl蛋白与各临床指标无明显相关性。结论重度子痫前期患者存在胎盘Axl蛋白及mRNA表达的异常;其胎盘Axl蛋白表达水平与疾病的某些重要临床指标相关。上述研究提示Axl的异常表达可能参与子痫前期的发病机制。Objective The aim of the present study was to investigate Axl expression in pregnant women with severe preeclampsia, and its role in the pathogenesis of preeclampsia. Mehtods Patients with severe preeclampsia and normal pregnancy were recruited from April 2012 to October 2012 in the department of Obstetrics and Gynecology, West China Second University Hospital, Sichuan University. The expressios of Axl protein and mRNA in placenta were detected by immunohistochemisty, Westen blotting, ELISA and real time PCR respectively. The results were analyzed by one-way ANOYA, Wileoxon and t-test, Pearson or Spearman's corre- lation coefficients. Results (1) The expression of Axl protein was found in maternal and fetal side of placental vascular endothelial cell in severe preeclampsia and normal pregnancy group. (2) There were no significant differences of Axl protein expression between maternal side and fetal side of the placenta in each group. (3) The expressions of Axl protein in placenta in severe preeclampsia group were significantly higher than that of in normal pregnancy (P〈0.05). (4) The expressions of Axl-mRNA in placenta in severe preeclampsia group were significantly lower than that of in normal pregnancy (P〈0.05). (5) In preeclampsia group, the Axl protein in placenta was positively correlated with blood pressure and 24h urinary protein, while negatively correlated with pregnancy gestational weeks. However, the Axl protein in placenta showed no significant correlation in normal pregnancy group. Conclusion Axl protein and mRNA expression in placentas of preeclampsia are found abnormal compared with normal pregnancy. In preeclampsia group, the expression of Axl protein in placenta is closely related with its important clinical parameters. Taken together, the abnormal expression of Axl might be involved in the pathogenesis of preeclampsia.
