摘 要:目的了解2010-2012年贵州省H3N2亚型流感病毒HA1基因变异特征。方法用MDCK细胞分离流感病毒,采用血凝抑制试验分型鉴定,选取23株H3N2亚型流感毒株进行RNA提取,逆转录扩增获得HA1基因产物,将其进行基因产物测序,测定的序列用相关的生物信息软件进行分析。结果贵州省2010年及2012年H3N2亚型流感病毒流行株与WHO疫苗推荐株A/Perth/16/2009(H3N2)亲缘关系较远,以至于H3亚型在我省成为优势株,引起广泛流行。结论 2010年及2012年H3N2亚型流感病毒流行株与WHO推荐的疫苗株之间的匹配性不完全一致。同时在2010-2012年间贵州省H3N2亚型流感病毒有持续的变异。Objective To understand characteristics of HA1 gene variation of H3N2 subtype influenza virus from 2010 to 2012 in Guizhou Province. Methods Influenza viruses isolated were cultured in MDCK cell and subtypes were tested by hemagglutination inhibition. There were 23 strains chosen randomly to be tested. Viral RNA was extracted and their HA1 genes were amplified by re- verse transcription polymerase chain reaction (RT--PCR). The PCR products were sequenced, and the gene sequences were ana- lyzed through biometic software. Results H3N2 influenza virus isolates in Gnizhon Province in 2010 and 2012 and vaccine strain A/Perth/16/2009 (H3N2) WHO recommended were distantly related, and H3 subtypes in Guizhou Province became the dominant strains, which caused be widespread. Conclusion The compatibility of H3N2 influenza virus isolates in 2010 and 2012 and vaccine strain WHO recommended are not completely consistent.At the same time, H3N2 subtypes of influenza A virus have continuous mutation between 2010-2012 in Guizhou Province.
