摘  要:目的研究有氧运动对中老年大鼠外周血内皮祖细胞增殖及黏附能力的影响。方法健康雌性2月龄和14月龄SD大鼠各12只,分为2月龄空白对照组,2月龄有氧运动组,14月龄空白对照组,14月龄有氧运动组4个组,每组6只。采用乳酸阈测定法确定有氧运动强度,以递增负荷跑台训练建立有氧运动模型,实验结束后12 h取大鼠外周血,采用密度梯度离心法分离单个核细胞,以EGM-2培养基对其体外培养,诱导分化获取内皮祖细胞,通过双荧光染色法观察细胞对乙酰化低密度脂蛋白的摄取和与荆豆凝集素1的结合以及形态学特征鉴定内皮祖细胞。倒置显微镜下计数贴壁细胞数量反映细胞增殖和黏附情况。结果与对照组相比,有氧运动组培养48 h和d 7的贴壁细胞数量都明显增加(P<0.01);内皮祖细胞黏附能力增强(P<0.01);14月龄大鼠的增殖和黏附能力均小于两月龄组(P<0.01)。结论有氧运动能促进中老年大鼠外周血中的内皮祖细胞增殖,且增加其黏附能力,但衰老则降低其增值和黏附。Objective To study the effects of aerobics on proliferative and adherent capacity in endothelial progenitor cells from peripheral blood of elderly rats. Methods 24 female Sprague-Dawley rats, including 12 ones of 2 months years old and the others of 14 months years old, were randomly and averagely divided into 4 groups such as control group with 2-month, aerobics group with 2-month, control group with 14-month and aerobics group with 14-month. Aerobic exercise intensity was defined by assay of lactic acid threshold, and incremental treadmill exercise was used to establish aerobics model. Mononuclear cells from peripheral blood were obtained by density gradient centrifugation 12 hours after the scheme has ended up. The ceils were suspended in conditioned medium EGM-2 for culturing in vitro. Their phenotypes were confirmed by uptake of acetylated LDL and binding of fluorescein isothiocyanate (FITC) -labeled Ulex europaeus agglutinin 1 (UEA-1) lectin. Inverted microscope observation was used to assay the morphological changes of endothelial progenitor ceils and cell count, so that the proliferation and adherence situation of endothelial progenitor cells were obtained. Results Adherent ceils in group aerobics increased more obviously than group control at 48h and day 7 (P^0.O1) and cell adhesion in group aerobics was better than control group (P〈0.01). Furthermore, both the proliferation and adhesion of EPCs in 14-month groups were worse than those in 2-month groups (P〈0.01). Conclusion Proliferative and adherent capacity of endothelial progenitor cells in elderly rats may be developed by aerobic exercise, on the contrary, they can be hindered by aging.
