摘  要:目的介孔二氧化硅纳米材料(mesoporous silica nanomaterials,MSNs)的理化性质如分散性会影响细胞摄取率,可能造成细胞凋亡。文中采用体外培养方法观察2种分散性不同的MSNs对293T细胞凋亡的影响。方法制备2种分散性不同的纳米材料MSN-1和MSN-2;透射电镜观察293T细胞对2种材料的吞噬;选择0、100、150、200和250μg/mL共5种浓度与293T细胞体外孵育24 h,流式细胞仪检测细胞凋亡反应。结果与MSN-1比较,MSN-2的分散性较好;293T细胞约吞噬10%的MSNs;与0μg/mL比较,100、150、200、250μg/mL的MSN-1与293T细胞共孵育引起细胞凋亡反应明显(P<0.05),而MSN-2并未引起明显的凋亡反应。结论在一定剂量下,分散性较佳的MSN-2在293T细胞内性能稳定,安全性较高。Objective Mesoporous silica nanomaterials(MSNs) plays a very important role in biomedical fields, its physico chemical properties such as dispersion can affect the cellular uptake rate, and maybe cause cell apoptosis. The aim of this study is to explain the apoptosis effect induced by MSNs with diverse dispersion in 293T cells in vitro. Methods Two MSNs named MSN-1 and MSN-2 with different dispersion were prepared; Transmission electron microscopy was used to observe the phagoeytosis of 293T cells caused by the two MSNs ; 293T cells were exposed to 0 μg/mL, 100 μg/mL, 150 μg/mL, 200 μg/mL and 250 μg/mL of MSNs in vitro for 24 h, and measured the apoptosis by flow cytometer. Results The dispersion of MSN-2 was better than MSN-1 ; about 10% MSNs were phagocyted by 293T cells ; MSN-1 incubated with 293T cells induced obvious apoptosis effect ( P 〈 0.05 ), while MSN-2 not. Conclusion MSN-2 with satisfactory dispersion was stable and safe in 293T cells in certain dose.
