摘 要:目的探讨二甲双胍对多囊卵巢综合征(PCOS)大鼠的促排卵作用及其对卵巢组织IL-6mRNA、TNF-αmRNA表达的影响。方法将32只雌性SD大鼠随机分为3组:空白对照组(A组,n=10),自由摄取食物和水31d;PCOS组(B组,n=12),来曲唑1mg/kg灌胃,21d后予以5mL/kg生理盐水灌胃10d;二甲双胍干预组(C组,n=10),来曲唑1mg/kg灌胃,21d后予以6%的二甲双胍5mL/kg灌胃10d。每日阴道涂片观察大鼠动情周期,末次灌胃24h后测定各组大鼠血清黄体生成素(LH)、雌二醇(E2)、睾酮(T)、胰岛素(INS)、空腹血糖水平,通过H-E染色观察卵巢形态,实时荧光定量PCR检测各组大鼠卵巢白细胞介素-6(IL-6)mRNA、肿瘤坏死因子-α(TNF-α)mRNA的相对表达。结果 C组较B组大鼠血清T、LH、INS明显降低(P<0.05),卵巢局部IL-6mRNA、TNF-αmRNA的表达下降(P<0.05),卵巢形态恢复正常。C组大鼠于用药的第6d全组恢复排卵,B组大鼠持续无排卵直至第10d,仅1只大鼠自发恢复排卵。结论二甲双胍能抑制卵巢局部IL-6mRNA、TNF-αmRNA的表达,有助于恢复卵巢形态,促进排卵,改善PCOS大鼠内分泌代谢紊乱。Objective To discuss the effects of Metformin on ovulation and expression of IL-6 mRNA and TNF-α mRNA in ovarian tissue of rats with PCOS . Methods Thirty-two female SD rats were randomly divided into 3 groups:normal group (group A , n= 10 ,allowed free access to water and standard rat diet) ,PCOS group(group B ,n=12 ,given an oral gavage dose of 1 mg/kg letrozole for 21 days ,then 5 mL/kg normal saline for 10 days) ,metformin group(group C ,n=10 ,administered through gavage letrozole with the dosage of 1 mg/kg for 21 days ,then the dosage of 5 mL/kg of 6% Metformin for 10 days) . Vaginal smears were taken daily to observe the estrous cycle of the female rats . Serum T , LH ,INS ,E2 ,FPG level were measured . Histological changes in ovarian tissues were observed with H-E staining . The relative expression of IL-6 mRNA and TNF-αmRNA on the ovaries of each group were detected by real-time PCR . Results Compared to the PCOS group ( P〈0 .05) ,the serum levels of T , LH ,INS decreased significantly in the metformin group . And ovarian morphology returned to normal with the significant decrease of IL-6 mRNA and TNF-αmRNA in ovarian tissue (P〈0 .05) . All the rats in the metformin group resumed ovulation by the sixth day of administration of metformin ,and the rats in the PCOS group except one remained anovulatory until the 10th day . Conclusion Metformin can be used for ovulation induction by decreasing the expression of IL-6 mRNA ,TNA-αmRNA on the ovaries and re-storing ovarian morphology . Metform can improve the endocrine and metabolic disorders in PCOS rats .
