摘 要:目的观察在大鼠窒息性心跳骤停模型中浅低温对脑线粒体膜电位的影响,从而进一步探索低温的保护机制。方法健康雄性Wistar大鼠15只,分为对照组(C组)、正常体温组(N组)和浅低温组(H组),每组5只。C组,仅进行麻醉气管插管及动静脉穿刺并呼吸机通气2 h;N组,窒息10 min后心肺复苏,复苏后体温维持(37±0.5)℃2 h;H组:复苏后体温维持在(33±0.5)℃2 h。各组于复苏后2 h断头取海马组织并提取海马线粒体,用JC-1荧光探针标记线粒体膜电位,用流式细胞仪检测荧光强度。结果 JC-1聚合物与单体的平均荧光强度比值在浅低温组(0.88±0.08)明显高于正常体温组(0.68±0.04,P<0.05),且均较对照组低(1.35±0.17,P<0.05)。结论在心跳骤停复苏后应用浅低温可以减少神经细胞线粒体膜电位下降从而保护脑I/R损伤。Objective To observe the effect of mild hypothermia on brain mitochondrial mem- brane potential in asphyxial cardiac arrest model in rats. Methods Fifteen male Wistar rats were randomly divided into 3 groups (n = 5 each) : control group ('C group) , normothermia grollp (N group) and mild hypothermia group (H group). Cardiac arrest model was produced by asphyxia. C group, anesthetized and ventilated for 2 h ; N group, asphyxia for 10 rain, rec- tum temperature was maintained at (37 ± 0.5 ) ℃ after resuscitation ; H group, "rectum temperature was maintained at (33 ±0.5)℃ after resuscitation. The animals were decapitated after 2 h of resuscitation and the mitochondria of hippocampus in each group was isolated. The mito-chondrial membrane potential was marked by fluorescent probe JC-1 and detected by flow cytometer. Results The fluorescence intensity ratio of aggregation and monomer in H group (0. 88 ±0. 08) was significantly higher than that in N group (0.68 ±0.04, P 〈0.05 ). Both of them were lower than in C group ( 1.35 ± 0. 17, P 〈 0. 05 ). Conclusion Mild hypothermia can maintain the mitochondria function by increasing neuron mitoehondrial membrane potential after ischemia-reperfusion (I/R). It maybe involved in the mechanism of protective effect of mild hypothermia on I/R injury.
