摘  要:目的研究载药氧化石墨烯对体外培养的大鼠胰岛活性及功能的影响。方法饱和氧化石墨烯吸附FTY720,观察吸附情况和缓释作用。采用不同浓度载药氧化石墨烯加入到胰岛培养基中,混合培养21天,采用SYTO-EB双染色法对各组培养3天、7天、14天、21天的胰岛细胞存活率和刺激分泌指数进行检测。结果氧化石墨烯饱和溶液的浓度为0.2 mg/mL,每毫升吸附FTY720 0.087 mg,载药率43.5%。与对照组相比较,实验组1中胰岛细胞存活率和分泌活性随着培养时间的延长明显下降(P<0.05),而实验组2和3与对照组相比无统计学差异(P>0.05)。结论氧化石墨烯可以吸附FTY720并能够缓释。载药石墨烯饱和溶液稀释2倍以上时对培养的胰岛细胞无明显毒副作用,作为潜在的药物载体可以安全应用于胰岛移植。Objective To study the effect of grapheme oxide loaded drugs on islet secreting activity and function in rats. Methods Saturated grapheme oxide FFY720 was absorbed with loaded, the adsorption and sustaining release were observed. Different concentration of grapheme oxide loaded drugs was added into islet medium for 21 d culture, then islet survival rate and stimulating secretion index were calculated at 3 d, 7 d, 14 d, 2l d with SYTO-EB staining. Results The concentration of saturated grapheme oxide was 0.2 mg/mL, and 0. 087 mg FTY720 was adsorbed by 1 mL saturated grapheme oxide, drug carrying rate was 43.5%. Compared with control group, the islet survival rate and secreting activity in experimental group 1 declined significantly with time growing ( P 〈 0.05 ) , while in experimental group 2 and 3, there was no significant difference (P 〉 0. 05 ). Conclusion Grapheme oxide (GO) can adsorb FTY720 and release it slowly. When saturated grapheme oxide loaded drugs was diluted for more than 2 times, it has no toxic and side effects on islets in vitro. Thus the concentration of grapheme oxide could be applied in islet transplantation safely as a drug carrier.
