摘  要:目的:探讨人参皂苷Rg1对脂多糖(LPS)诱导心肌细胞肥大的保护作用及机制。方法:利用体外培养模型,以脂多糖1 mg/L诱导心肌细胞肥大,观察不同剂量人参皂苷Rg1(12.5、25、50μmol/L)及环孢菌素A(CsA)对脂多糖诱导的心肌肥大的抑制作用。用考马斯亮蓝法测心肌细胞蛋白质含量;消化分离法及计算机图像分析系统检测心肌细胞体积;倒置相差显微镜下观察细胞形态大小;采用Fluo-3/Am荧光染色剂负载细胞,在激光共聚焦显微镜下测定细胞内钙离子浓度的瞬间变化;电泳法测定钙调磷酸酶(CaN)的表达。结果:与正常对照组相比,脂多糖模型组使心肌细胞总蛋白质含量增加,细胞体积增大,心肌细胞内[Ca2+]i瞬间峰值增大,钙调磷酸酶表达增加。与模型组相比,人参皂苷Rg1(25、50μmol/L)可有效改善脂多糖诱导的心肌细胞肥大,抑制心肌细胞内[Ca2+]i瞬间峰值增大,抑制钙调磷酸酶表达增加。结论:人参皂苷Rg1可通过抑制Ca2+/钙调磷酸酶信号通路抑制脂多糖诱导的心肌细胞肥大。Objective: The aim of the prcsent study was to study the protective effect of Ginsenoside Rgl on LPS-induced myocardial hypertrophy, and to explore a possible mechanism. Methods: The hypertrophic myocytes were induced by lipopolysaccharide (LPS) 1 mg/L and then dif- ferent doses of Ginsenoside Rgl ( 12.5,25,50μmol/L) and CsA were administered. The total protein content was measured by the method of Bradford. The cardiomyocytes size was measured by computer photograph analysis system. Morphologic changes of cardiomyocytes were ob- served with inverted phase contrast microscope; [ Ca2+] i transient was measured by Laser scanning confocal microscopy by cell-loading Fluo- 3/Am. The expression of CaN was determined by Western blot. Results: LPS 1 mg/L increased the total protein content, the cardiomyocytes size, [ Ca2+ ]i transient, and the expression of CaN. Compared with the model group, The total protein content, cell size were decreased, [ Ca2+ ]i transient and the increased expression of CaN were decreased too in Ginseneside Rgl (25,50μmol/L) and CsA group. Conclusion: Ginsenoside Rgl has a protective effect on LPS-induced myocardial hypertrophy, which is partially via Ca2+ and CaN signling pathway.
