不同桂枝配伍对CIA大鼠血清TNF-α、IL-1β、IL-6因子的影响

摘　　要：目的:观察桂枝配伍对类风湿性关节炎模型促炎细胞因子(PICK)的调控作用。方法:以Ⅱ型胶原(CⅡ)诱导的免疫性关节炎(CIA)大鼠为RA动物模型,通过关节炎指数观察桂枝、桂枝配伍附子、桂枝配伍川乌、桂枝配伍白芍及桂枝配伍当归组的治疗效果;采用ELISA法测定大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)活性。结果:模型组大鼠血清TNF-α、IL-1β及IL-6含量明显升高;经治疗后桂枝及各配伍组关节炎指数明显均降低;与模型组比较,桂枝组(1.8g/kg)明显降低TNF-α(P<0.05),桂枝白芍组(3.6g/kg)明显降低TNF-α、IL-1β、IL-6(P<0.01或P<0.05),桂枝附子组(4.5g/kg)、桂枝川乌组(4.5g/kg)及桂枝当归组(3.6g/kg)明显降低TNF-α、IL-6(P<0.01或P<0.05)。结论:桂枝及不同配伍可以降低CIA大鼠血清中异常增高的TNF-α、IL-6浓度,提示这可能是其治疗RA的作用机理之一。Objective： To observe the the effect on proinflammatory cytokine in the serum of rheumatoid arthritis model by the treatment of Cassia twig compatibility environment. Methods： Arthritis of rat induced by type II collage was used as rheumatoid arthritis model. Selling of rats arthritis index was assessed to observe the therapeutic effect of Cassia twig,Cassia twig with Monkshood,Cassia twig with Radix Aconiti, Cassia twig with Radix Paeoniae alba or Cassia twig with Angelica sinensis. The activity of TNF-α, IL-1β and IL-6 in serum was investigated with ELISA method. Results： The activity of TNF-α, IL-1β and IL-6 in model group were significant higher than the normal group. Compared with that of model group, Cassia twig and the compatibility groups arthritis index in rats after treatment was significantly alleviated. The Cassia twig group（1.8g/kg） significantly reduces TNF-α（ P 〈 0.05 ）, Cassia twig and Radix Paeoniae alba group（3.6g/kg） significantly reduces TNF- α,IL-1β,IL-6（P 〈0.01 或 P 〈0.05）, Cassia twig and Angelica sinensis group（3.6g/kg） ,Cassia twig and Radix Aconiti group（4.5g/kg）, Cassia twig and Monkshood group（4.5g/kg） significantly reduces TNF-α,IL-6 （ P 〈 0.01 或 P 〈 0.05 ） compared with the model group. Conclusion： Cassia twig compatibility environment has an effect on immune arthritis induced by type II collage, and the decrease of TNF-α and IL-6 in serum may be one of its mechanisms.
