复方抗毒升白合剂的抗肿瘤活性及其机制探讨

摘　　要：目的：初步探讨复方抗毒升白合剂的体内外抗肿瘤活性及其作用机制。方法：MTF法测定抗毒升白合剂对人肝癌细胞BEL7404和HepG2的抑制作用；S180肉瘤模型小鼠灌胃给予抗毒升白合剂，计算肿瘤抑制率和免疫器官指数，制作肿瘤、肝脏及脾脏切片；Elisa方法测定荷瘤小鼠血清IL-2、Bcl-2及wtp53含量，探讨抗毒升白合剂的抗肿瘤机制。结果：抗毒升白各剂量组（15．63～1000μg/ml）对人肝癌BEL7404及HepG2细胞均表现出一定的抑制作用；各剂量（10、20、40g/kg）对S180肉瘤模型小鼠肿瘤抑制率分别为49．03％、44．78％和32．78％，给药组免疫器官指数均高于环磷酰胺组；抗毒升白合剂能降低荷瘤小鼠血清Bel-2含量，升高IL-2及wtp53的含量。结论：抗毒升白合剂具有一定的抗肿瘤作用，其作用机理可能是通过降低荷瘤小鼠Bcl-2的含量，升高荷瘤小鼠IL-2及wtp53的含量来抑制肿瘤的生长。Objective：To exam the anti-tumor activity of compound Kangdushengbaiheji and explore the possibility of anti-tumor mechanism. Methods： The inhibitory rate of tumor cells BEL7404, HepG2 with Kangdushengbaiheji were detected by MTT in the vitro anti-tumor experiments. Mice with S180 administrated Kangdushengbaiheji ,the tumor inhibition rate and immune organ index were calculated, and the tumor, liver and spleen slice were made;The content of IL-2 ,Bcl-2 and wtp53 in the serum were measured by Elisa to investigate the anti-tumor mechanism. Results：Kangdushengbaiheji （15.63 - 1000μg/ml）showed some inhibitionon liver cancer cell Bel-7404 and HepG-2 ;The tumor-inhibitory rates of low, middle and high dose （20,30,40g/kg） of Kangdushengbaiheji groups to S180 were 32.78% ,44.88% and 49.03%, and the immune organ index of Kangdushengbaiheji groups were higher than cyclophosphamidum group ; Kangdushengbaiheji could reduce the content of Bcl-2,increase IL-2 and wtp53 in tumor-beating mice. Conelusion：Kangdushengbaiheji have anti-tumor activity. The inhibiting effects on tumor may be related to increasing the p53 protein and IL-2 protein and decreasing the Bcl-2 protein expression of mice.
