摘 要:目的:探讨丹莪妇康煎膏对子宫内膜异位症大鼠异位子宫内膜组织中血管内皮生长因子(VEGF)mRNA表达和肿瘤坏死因子(TNF-α)表达的影响。方法:采用自体移植法建立子宫内膜异位症大鼠模型,将造模成功的大鼠随机分为模型组、孕三烯酮组(1.25mg/kg)、丹莪妇康煎膏高剂量组(6g/kg)、低剂量组(3g/kg),另设假手术组。给药30天后,观察异位病灶外观及形态学改变,以RT-PCR法检测大鼠在位以及异位内膜组织中VEGF mRNA表达,以免疫组化法检测TNF-α的表达。结果:与模型组相比,丹莪妇康煎膏高剂量(6g/kg)、低剂量(3g/kg)组大鼠子宫内膜移植物体积均显著缩小,异位内膜组织出现萎缩,异位内膜组织中VEGF mRNA的表达显著降低,丹莪妇康煎膏高剂量组(6g/kg)大鼠异位内膜组织中TNF-α的表达显著降低,低剂量组(3g/kg)TNF-α的表达与模型组相比虽无显著性差异,但也有降低趋势。结论:丹莪妇康煎膏通过降低VEGF和TNF-α的表达,抑制子宫内膜细胞的侵袭和增殖能力,这可能是其治疗子宫内膜异位症的作用机制之一。Aim: The purpose of the study was to observe the effects of Dane Fukang Decocted Extract(DFD) on the expression level of vascular endothelial growth factor (VEGF) and tumor necrosis factor(TNF-α) in endometriosis(EMS) rats. Methods: The model of EMS rats were made by implanted tunica intima method. Then the rats were randomly divided into 5 groups, sham operation group, EMS model group, Gestrinone group( 1.25mg/kg), high dose (6g/kg) and low dose (3g/kg) DFD group. After continously intragastric administration for 30 days, the morphological changes of ectopic endometrium tissue were observed, the expression level of VEGF mRNA was detected by RT-PCR, and the expression level of TNF-α was detected by immunochemical methods. Results: Compared with EMS model group, high dose (6g/kg) and low dose(3g/kg) DFD could inhibit the growth of ectopic endometrium tissue, significantly down-regulate the expression level of VEGF mRNA( P 〈 0.01 ). High dose DFD(6g/kg) could significantly down-regulate the expression level of TNF-α( P 〈 0.01 ). But there was no notable difference of the expression level of TNF-α between low dose DFD group(3g/kg) and EMS model group( P 〉 0.05). Conclusion: DFD could treat EMS through down-regulating the expression levels of VEGF and TNF-α.
