摘 要:目的:检测一次急性力竭运动后大鼠肝扭转原肠胚形成同系物1(Twisted gastrulation,TWSG1)、SMAD4(drosophila mothers against decapentaplegic protein)和肝铁贮存的动态变化,了解急性运动对铁代谢的调控作用。方法:雄性SD大鼠30只(220±10)g分为对照组(CG组,6只)和运动组(EG组,24只),运动组再分为E0h组、E3h组、E6h组和E24h组。运动组进行跑速为30 m/min的一次急性力竭跑台运动。分别于运动结束后0 h、3 h、6 h和24 h采用RT-PCR法测定大鼠肝脏TWSG1、骨形态发生蛋白6(Bone morphogenetic protein6,BMP6)和SMAD4 mRNA表达,采用试剂盒检测肝非血红素铁。结果:(1)E3h组TWSG1 mRNA表达量显著低于CG组和E24h组(P<0.01),E24h组显著高于其它各组(P<0.01)。E3h组BMP6和SMAD4 mRNA显著高于其它各组(P<0.01),E24h组显著低于其它各组(P<0.01)。(2)E3h组肝非血红素铁与CG组比较显著下降(P<0.01)。结论:一次急性力竭运动中TWSG1、BMP6及SMAD4通路对机体铁状态发挥重要调控作用。Objective To investigate the dynamic change in expression of twisted gastrulation (TWSG1) and drosophila mothers against decapentaplegic protein (SMAD4)after an acute exercise and the regulatory effect of the exercise on iron metabolism. Methods 30 male SD rats (220 g±10 g)were randomly divided into the control group(C, n = 6)and the exercise group(E, n = 24, nmning at the speed of 30m/min until exhaus- tion). The hepatic TWSG1, BMP6 (Bone morphogenetic protein6), and SMAD4 mRNAs were examined by RT-PCR immediately (E0h), 3 hours ( E3h), 6 hours (E6h), and 24 hours (E24h) after exercise. Hepatic non- heme iron was determined by Iron Kits. Results ( 1 ) The expression of TWSG1 mRNA in group E3h de- creased significantly compared with that in groups C and E24h(P 〈 0.01 )and in group E24h increased signif- icantly compared with that in other groups (P 〈 0.01 ). The expression of TWSG1 mRNA in group E3h de- creased significantly compared with that in groups C and E24h (P 〈 0.01 ). The expression of TWSG1 mRNA in group E3h significantly increased compared with that in other groups(P 〈 0.01 )and in group E24h decreased significantly compared with that in other groups (P 〈 0.01 ). (2)The liver non-heme iron in group E3h decreased significantly compared with that in other groups (P 〈 0.01 ). Conclusion The TWSG1, BMP6 and SMAD4 pathway might play an important role in regulating iron metabolism during acute exhaustive ex- ercise.
