摘 要:目的探讨高原红细胞增多症(HAPC)患者外周血单个核细胞(PBMC)miR-210和caspase-8 mRNA表达的变化。方法采集高原红细胞增多症患者(53例)以及健康个体(46例)的外周静脉血样,以实时荧光定量PCR检测PBMC miR-210和caspase-8 mRNA的表达。结果与对照组比较,HAPC患者PBMC miR-210的表达增高,caspase-8 mRNA的表达降低,差异有统计学意义(P<0.01);HAPC患者PMBCmiR-210和caspase-8mRNA的表达呈负相关(r=-0.792,P<0.01)。结论 HAPC的发生可能与miR-210的表达升高和caspase-8 mRNA的表达降低有关。Objective To discuss the clinical significance of the expressions of microRNA-210 and its target gene easpase-8 mRNA in peripheral blood mononuclear cells (PMBC) of patients with high altitude polyeythemia (HAPC). Methods A total of 53 patients with high altitude polyeythemia (HAPC) and 46 healthy people were selected and the expressions of microRNA-210 and caspase-8 mRNA in PMBC were examined by the real-time quantitative PCR. Results Compared with the control, the expression of mieroRNA-210 up-regulated significantly (P〈0.01) and the expression of caspase-8 mRNA in patients with HAPC decreased obviously (P〈0.01). The expression of microRNA-210 in PMBC of patients with HAPC was negatively correlated with the expression of easpase-8 mRNA (P〈0.01). Conclusion The abnormal expressions of mieroRNA- 210 and easpase-8 mRNA may be involved in the pathogenesis of HAPC.
