摘 要:目的:观察肾衰养真胶囊(Shenshuai Yangzhen Capsule,SYC)对慢性肾衰竭(Chronic renal failure,CRF)营养不良大鼠下丘脑信号传导转录活化因子3(Signal transducer and activator of transcription3,STAT3)表达的影响。方法:采用含0.5%腺嘌呤及4%酪蛋白饲料联合喂养的方法制作CRF营养不良大鼠模型,检测BUN、Scr、Hb,Alb等生化指标,观察其营养不良发生时间,符合CRF营养不良模型的,随机分为正常组、模型组、SYC组和开同组。治疗4周后,应用放射免疫法检测血浆瘦素(Leptin,LP)水平;Western免疫印迹法检测下丘脑STAT3蛋白表达水平。结果:治疗后,SYC组大鼠营养状况明显改善Hb、Alb显著升高,血浆LP及下丘脑STAT3蛋白表达显著下调。结论:SYC可改善CRF营养不良大鼠的营养状况,其机制可能是通过下调血浆LP水平及其信号转导蛋白STAT3表达来实现的。Objective:To investigate the effect of Shenshuai Yangzhen capsule (SYC) on hypothalamic signal transducer and activator of transcription3 ( STAT3 ) in chronic renal failure (CRF) rats with malnutrition (MN). Methods : Fifty - two male SD rats of SPF grade were established into CRF - MN model by feeding with 0. 5% adenine and 4% easelnum. The happening time of MN in them was recorded. Successfully modeled rats were randomized into CRF control group, Ketosteril group, SYC group. Besides, a normal control group was set with 8 healthy rats. After being treated for 4 weeks, blood leptin levels in rats were determined by radioimmunoassay;expressions of STAT3 protein were detected by Western blot. Meantime, the renal fmwtion related indexes, including albumin (ALB) and haemoglobin ( Hb), were measured. Results : CRF rats revealed MN at the end of 8th week after modeling. Compared with CRF control group, the condition of MN in SYC group was significantly improved,showing increase of ALB and Hb (P 〈 0.01 ) and decrease of LP levels in plasma (P 〈 0. 05) and the SOCS3 expression was down - regulated in hypothalamus (P 〈 0. 05). Conclusion:SYC can improve the MN condition in rats with CRF, which is possible by way of depressing LP levels in plasma and downregulating SOCS3 expression in hypothalamus.
