阳和化岩汤对大鼠乳腺癌癌前病变的抑制作用研究

摘　　要：目的:建立大鼠乳腺癌癌前病变疾病模型,研究阳和化岩汤对大鼠乳腺癌癌前病变的抑制作用。方法:采用DMBA诱导建立大鼠乳腺癌癌前病变模型,9周后分组用药干预治疗4周,观察大鼠体重增长及肿瘤出现情况,取乳腺组织观察病理组织学形态。结果:造模组大鼠肉眼肿瘤发生率92.86%,平均肿瘤个数为4.21,癌前病变和浸润性癌发生率分别21.43%、71.43%,与空白对照组比较具有显著差异(P<0.01)。各治疗组大鼠肉眼肿瘤发生率及平均肿瘤个数与疾病模型组比较,均明显降低和减少(P<0.05);阳和化岩汤组肿瘤发生率为64.71%,平均肿瘤个数为1.82,与其他治疗组比较,无统计学差异(P>0.05)。各治疗组大鼠乳腺组织病理形态学较疾病模型组病变程度轻(P<0.05);阳和化岩汤组癌前病变发生率为47.06%,浸润性癌发生率为29.41%,与其他治疗组比较,无统计学差异(P>0.05)。结论:DMBA小剂量分次给药建立大鼠乳腺癌癌前病变动物模型具有明显优越性,阳和化岩汤为代表的温阳散结中药能够部分抑制乳腺癌癌变,适于乳腺癌高危人群的预防治疗。Objective： To establish the rat breast precancerous lesion disease models ,and to research Yanghe Huayan Decoction inhibiting breast cancer premalignant lesion of rats. Methods： Rat mammary precancerous models were established by DMBA. 9 weeks later,the rats were grouped and treated for 4 weeks separately, observing rats about weight growth,tumor occurrence, and pathological morphology of breast tissue. Results： In model- group rats, the tumor incidence rate was 92.86% ,the average number of tumor was 4.21, and the precancerous lesions and invasive cancer rates were respectively 21.43% ,71.43% ,and showed significant different compared with the control -group （ P 〈 0.01 ）. The tumor incidence rate and average number of tumor in each treated group were significantly reduced, compared with the model group （ P 〈0.05）. In the Yanghe Huayan Decoction group,the incidence rate of tumor was 64.71% ,the average number of tumor was 1.82, and showed no significant difference compared with the other treated groups （ P 〉 0.05 ）. In pathological morphology severity,each treatment group was lighter than the model group （ P 〈 0.05 ）. In the Yanghe Huayan Decoction group,the precancerous incidence rate was 47.06% ,invasive cancer incidence rate was 29.41%, and showed no significant difference compared with the other treatment groups （ P 〉 0.05）. Conclusion： It was superior that DMBA was afforded in small dose and repeatedly to establish rat breast precancerous lesioils models. And warming yang and dispersing phlegm method delegated by Yanghe Huayan Decoction could inhibit breast cancer carcinogenesis in a certain distant, and should be suitable for the prevention and treatment of breast cancer high - risk groups.
