云南白药胶囊对C518大鼠膝关节退变软骨细胞增殖和COL-Ⅱ、MMP-13表达的影响

摘　　要：目的:检测云南白药胶囊对C518大鼠膝关节退变软骨细胞增殖和COL-Ⅱ、MMP-13表达的影响。方法:用MTT法检测云南白药胶囊对C518大鼠膝关节退变软骨细胞增殖的影响,并对云南白药胶囊干预后的C518大鼠膝关节退变软骨细胞进行COL-Ⅱ、MMP-13免疫组化染色,同时观察其干预效果。结果:在药物对细胞增殖的影响上,在同一时间点的不同浓度中,各组之间都有显著性差异(P<0.01)。在同一时间点的云南白药组、氨糖美辛组中,各浓度之间都有显著性差异(P<0.01)。在生理盐水组中,各浓度之间都无显著性差异(P>0.05)。在COL-Ⅱ、MMP-13表达水平上,各组之间都有显著性差异(P<0.01)。结论:云南白药胶囊对C518大鼠膝关节退变软骨细胞有促进增殖的作用,并且细胞对药物浓度有依赖作用。云南白药胶囊能调节C518大鼠膝关节退变软骨细胞中COL-Ⅱ、MMP-13的表达水平,从而延缓KOA的病变发展。Objective： To detect the Yun-nan-bai-yao capsule influence to the degenerative callilage cells of C518 rat＇s knee joint in proliferation and COL-Ⅱ , MMP-13 expression. Methods： We used MTT to detect the effect of Yun-nan-bai-yao on the prolif- eralinn of degenerative cafilage cells, which were frnm C518 rat＇s knee joint, then we used COL-Ⅱ , MMP-13 in dye the degenera- tive cartilage cells of C518 rat＇s knee joint, which were intervened with Ytmnan Baiyao, and observed the intervention effect. Results： From the effects of drugs on cell proliferation, there were significant differences beween the grnups with different concentration at the same time （P 〈 O. Ol ） . At the same point of time, Yunnan Baiyao capsule grnup and Osaminethacine gr^mp had significant difference with different cnneentratinn （P 〈0. Ol ） . In physinlogical saline grnups, there were nnt signiflcanl differences between different con- centration （ P 〉 0. 05） . In the expression level of COL--Ⅱ and MMP-13, there were significant differences between the grups （ P 〈0. 01 ） . Conclusion： Yun-nan-bai-yao capsule can promote the proliferatinn of degenerative eartilage cells, which are from C518 rat＇ s knee joint, and the cells depended nn the drug concentration. ~un-nan-bai-yao capsule can regulate the expression level of COL--Ⅱ ,MMP-13 in degenerative cartilage cells, which are ft^om C518 rat＇ s knee joinl, /hereby delaying tile de^elopmen/of KOA.
