摘 要:该课题对胚胎期12.5、13.5、14.5d的C57/BL6孕鼠胎鼠以不同的电压参数进行子宫内电穿孔,分析比较不同电压对不同胚胎期胎鼠的存活率,从而总结出适应胚胎期12.5、13.5、14.5d胎鼠的最优电转电压参数。结果显示,胚胎早期,电转所需要的电压相对小一些,同样胚胎早期的胎鼠的存活率低一些。。在E14.5胎鼠脑室单侧电穿孔电转带有PCDF.EGFP质粒。该质粒表达绿色荧光蛋白(GFP)。进一步分析了电转后不同天数转染细胞的迁移程度,结果显示,在电转后2d,转染阳性的细胞主要分布在脑室和中间带,而在转染后3d,转染阳性细胞在脑室、中间带及皮层均有分布。继而对电转后的皮层厚度进行了分析,结果显示,电转皮层的厚度与未进行电转的皮层没有明显区别。该结果为子宫内电转在皮层发育中的应用提供了技术支持,并表明子宫内电转可以应用在皮层发育研究中。The subjects for embryonic stage 12.5, 13.5, 14.5 the C57/BL6 pregnant mouse fetal to different parameters utero electroporation were analyzed with different voltage of different embryonic period of fetal mouse survival rate,thus to summarize for embryonic day 12.5, 13.5, 14.5,the optimal voltage parameters of fetal mice.The results show that the early embryo,electrical voltage is required relatively small and early embryonic fetal mice survival rate is low.El4.5 fetal mouse ventricular unilateral electroporation transfer with the PCDF-EGFP plasmid,the plasmid expressing green fluorescent protein(GFP).We further analyzed the electrically different days after transfection cells transfected migration degree.The results showed that 2 days after electroporation,transfection-positive cells are mainly distributed in the ventricle and the intermediate zone,3 days after transfection,the transfected positive cells were distributed in the ventricle,the intermediate zone and cortex.We then analyzed cortical thickness.The results show that the electrical cortical thickness is no significant difference.Our results for cortical development in utero electroporation in providing technical support show that in utero electroporation can be applied to the study of cortical development.
