高效液相梯度洗脱法测定尿中氢化可的松和可的松含量

摘　　要：目的 :测定尿中氢化可的松与可的松的比值以表达人体肾脏 11β 羟化类固醇脱氢酶活性 ,为其研究提供一个准确、精密、重现性好的分析手段。方法 :HPLC UV法 ,梯度洗脱 ,内标定量。结果 :尿中氢化可的松和可的松在 10～ 10 0 .0ng·ml-1范围内其线性回归方程分别为Y =0 .0 1X + 0 .0 2 7和Y =0 .0 12X + 0 .0 2 5 ,氢化可的松和可的松相对回收率分别为 (10 5 .8±3 .1) %和 (96.6± 3 .6) % (n =15 ,RSD为 2 .4 0 %～ 4 .87% )。 3 4例健康成人尿液氢化可的松和可的松浓度分别为 (2 5 .2±14 .3 )和 (4 6.2± 2 6.7)ng·ml-1,氢化可的松与可的松的比值为 (0 .5 9± 0 .18)。结论 :本法灵敏可靠 。OBJECTIVE:To investigate 11β hydroxystroid dehydrogenase activity,an exact precision reproducible assay was developed for measuring the ratio of cortisol and cortisone.METHODS:Gradient HPLC was used.RESULTS:The linear regress equations for cortisol and cortisone were Y = 0.01 X - 0.027 and Y = 0.012 X - 0.025 ,the percentages of their average recovery were ( 105.8 ± 3.1 )% and ( 99.6 ± 3.6 )%( n =15, RSD : 2.40% ～ 4.87% ) respectively.CONCLUSIONS:This method is able to satisfy demands for measuring glucocorticoid of human urine,and it is very sensitive and credible.The concentration of cortisol and cortisone in 34 healthy volunteers's urine were ( 25.2 ± 14.3 )ng·ml -1 and 46.2 ± 26.7 ng·ml -1 and the ratio of cortisol and cortisone was 0.59 ± 0.18 .
