小麦株高近等基因系的RAPD标记研究

摘　　要：采用RAPD技术 ,以 4个小麦株高基因近等基因系 (分别含有rht、Rht1、Rht2、Rht3)为材料 ,对 2 96个单一随机引物 (10个核苷酸 )进行了筛选。发现 2 5个引物的扩增产物在近等基因系间表现出特异性。在 6次重复试验中 ,有 18个引物的特异扩增片段不能重复 ,6个引物可以重复 2～ 3次 ,唯有OPAM 0 1在全部试验中均能稳定重复 ,其特异扩增片段OPAM 0 1186 0 可以作为Four near isogenic lines which contained rht,Rht1,Rht2,Rht3 respectively were used for polymorphic analysis at molecular level by RAPD technique. Out of 296 10 mer oligonucleotide primers screened, 25 showed polymorphism among near isogenic lines. During 6 repeat tests of these primers, 18 could not give the repeat results, 6 could repeat 2 to 3 times, only primer OPAM01 showed the stable results in which a 1.86 kb additional band was found in rht line. OPAM01 1860 could be identified as a RAPD marker linked to rht gene in wheat.
