清热排毒胶囊对急性痛风大鼠模型IL-1β、IL-8和TNF-α的影响

摘　　要：目的:研究清热排毒胶囊对尿酸钠(MSU)诱导的急性痛风性关节炎模型大鼠踝关节浸出液中IL-1β、IL-8和TNF-α的影响,探讨其治疗急性痛风性关节炎的部分作用机理。方法:选用健康SPF级SD雄性大鼠60只,随机分成清热排毒胶囊高、中、低剂量组,双氯芬酸钠缓释片组(戴芬组)、模型组、空白组。各组给予相应药物灌胃7天,于第5天灌胃给药1h后予尿酸钠晶体局部关节腔注射形成大鼠急性痛风性关节炎模型。测量大鼠踝关节关节肿胀度,应用ELISA法测定踝关节浸出液中IL-1β、IL-8和TNF-α的含量。结果:模型组大鼠关节肿胀明显,踝关节浸出液中IL-1β、IL-8和TNF-α含量明显高于正常组(P<0.01);清热排毒胶囊中剂量组和戴芬组大鼠关节肿胀度降低明显,IL-1β、IL-8和TNF-α的含量明显低于模型组(P<0.01,P<0.05);清热排毒胶囊中剂量组中IL-1β、TNF-α表达水平低于戴芬组(P>0.05)。结论:清热排毒胶囊对急性痛风性关节炎具有一定的治疗作用,其发挥抗炎作用机制可能与抑制滑膜组织中IL-1β、IL-8和TNF-α的表达有关。Objective： To observe the effects of Qingrepaidu Capsule on IL-1, IL-8 and TNF- α in the joint leachate of acute gouty arthritis induced by MSU in rats, and explore its mechanism of action on treating acute gouty arthritis.Methods： SPF 60 healthy male SD rats were randomLy divided into 6 groups： normal group, model group, Qingrepaidu capsule high, middle and low dose group, Diclofenac sodium group.Each group were given corresponding drug for 7 days.Rat gouty arthritis model were established by monosodium urate crystal intraarticular injection in 5 days after lavage administration for 1 hour.Measured the joint swell degree of right ankle joint, and the expression level of IL-1 β , IL-8, TNF- α in the joint leachate by ELISA assay. Results ： In the rat of the model group, the joint swelling were very obviously.The contents of IL-1 β , IL-8, TNF- α in the joint leachate of the model group were significantly higher than that of the normal group with a significant difference （ P〈0.01 ） ; Qingrepaidu capsule middle dose group and Diclofenac sodium group can significantly reduce the degree of ankle swelling, the contents of IL-1 β , IL-8, TNF- α were significantly decreased with a significantly difference compared with the model group （ P〈0.01, P〈0.05 ） ; The level of IL-1β and TNF- α in the joint leachate of Qingrepaidu capsule middle dose group lower than Diclofenac sodium group, there was no significant difference between two groups （ P〉0.05 ） .Conclusion： Qingrepaidu Capsule is effective on treating acute gouty arthritis.Its mechanism of anti-inflammatory effects could be related to inhibit the expression of IL-1 β , IL-8, TNF- α in synovial tissue of rats.
