“六郁同治”法对动脉粥样硬化易损斑块ApoE-/-模型小鼠脂代谢及高敏C反应蛋白的影响  被引量:5

摘  要:目的观察"六郁同治"法对动脉粥样硬化易损斑块ApoE-/-模型小鼠血脂及高敏C反应蛋白(hsCRP)表达的影响,探讨其稳定易损斑块的作用机制。方法将6周龄ApoE-/-小鼠高脂喂养24周后随机分为中药高剂量组、中药低剂量组、辛伐他汀组、模型组,治疗12周后检测其血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、hsCRP水平,并观察斑块病理形态。结果光镜下显示造模小鼠主动脉管壁厚薄不均,可见由大量泡沫细胞及胆固醇结晶形成的粥样斑块期病灶,表示造模成功。与模型组比较,中药低剂量组、中药高剂量组、辛伐他汀组血清TC、TG、LDL-C水平明显降低(P<0.05),HDL-C水平明显升高(P<0.05),斑块内脂质成分减少(P<0.01),hsCRP表达水平降低(P<0.01)。结论 "六郁同治"法可稳定易损斑块,延缓动脉粥样硬化进程,其作用机制可能与抑制血脂代谢及炎性因子hsCRP表达有关。Objective Through establishing vulnerable plaque(VP) of atherosclerosis ApoE-/-mice model, to observe the effect of 'treating six yu together' method on expression of blood lipid and high-sensitivity C-reactive protein(hsCRP) of VP model, so as to explore its mechanism on stabilizing VP. Methods 6-month-old ApoE-/-mice after 24 weeks of high fat diet were randomly divided into a high-dose TCM group, low-dose TCM group, simvastatin group and model group, the level of serum total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterin (LDL-C), high density lipoprotein cholesterin (HDL-C) and hsCRP was detected after 12 weeks of treatment, and pathomorphism of plaque was observed. Results The light microscope showed non-uniform thickness of artery wall of the model mice and atherosclerosis plaque focus, which meant the model was established successfully; compared to the model group, the level of TC, TG and LDL-C in the other groups was decreased significantly(P 〈0. 05), while the level of HDL-C was increased obviously (P 〈 0. 05), intramyocellular lipid composition of plaque was reduced ( P 〈 0.01 ), and the level of hsCRP was lowered(P 〈0. 01 ). Conclusion The method of ' treating six yu together' could stabilize the VP and delay the progress of atherosclerosis, and its mechanism might be related with restraining the blood lipid metabolism and the expression of inflammatory factor hsCRP.
