摘  要:目的:探讨S期激酶相关蛋白2(S-phase kinase-associated protein 2,Skp2)在肺癌患者外周血中微转移中的诊断价值。方法:选取40例在本院就诊经病理组织学检查确诊为非小细胞肺癌(NSCLC)的患者作为肺癌组,选择同期诊断为肺部良性疾病的20例患者为肺部良性疾病组。未进行任何治疗前收集各研究对象的外周血,实时荧光定量PCR方法检测血Skp2mRNA并进行评价分析。结果:(1)肺癌组外周血中Skp2mRNA相对表达量明显高于肺部良性疾病组,差异具有统计学意义(P=0.006);(2)肺癌组外周血中Skp2mRNA的阳性表达率分别为77.5%,肺部良性疾病组阳性表达率为8%,明显低于肺癌组,差异具有统计学意义(P<0.01);(3)肺癌组外周血中Skp2mRNA的表达与患者肿瘤的TNM分期显著相关(P=0.004),与年龄、性别、吸烟、肿瘤的组织学类型及分化程度无明显相关性。结论:外周血Skp2mRNA表达可作为肺癌细胞的分子标记物用于NSCLC临床诊断。Objective=To analyze the clinical significance of expression of S-phase kinase-associated protein 2 mRNA in peripheral blood of patients with lung cancer. Methods= Real-time fluorescence quanti- tative RT-PCR was applied to investigate the quantitative expression of Skp2 mRNA in peripheral blood of 40 patients with lung cancer and 20 patients with benign lung diseases in our hospital during the same peri- od. Results- (1) The mRNA expression levels of Skp2 in NSCLC patients were significantly higher than those in benign lung disease patients (P=0. 006). (2) The positive rates of mRNA expression of Skp2 in peripheral blood from 40 patients with NSCLC was 77.5%, while the positive rates of Skp2 expression in peripheral blood from patients with benign lung diseases was 8%, which was significant lower than those of patients with NSCLC (P〈0.01). (3) For patients with NSCLC, the tumor TNM stage were significantly correlated with positive rates of Skp2 mRNA expression (P=0.01), whereas tions between positive rates of Skp2 mRNA expression and age, gender, smoking, there were no correla histopathlogieal type and tumor cell differenting degrees. Conclusions. The Skp2 mRNA expressions in peripheral blood could be used as the molecular markers for diagnosis of NSCLC.
