二氯乙酸钠诱导肺腺癌细胞株A549凋亡的实验研究

摘　　要：目的研究小分子药物二氯乙酸钠(DCA-Na)对人肺腺癌细胞株A549体外抗肿瘤及其诱导肿瘤细胞凋亡的作用,探讨其抗肿瘤作用的可能机制。方法采用四甲基偶氮唑盐(MTT)法观察A549细胞在不同浓度DCA-Na处理24、48、72 h后生长受抑制情况;使用流式细胞仪、透射电镜和Caspase 3试剂盒检测观察DCA-Na对A549细胞凋亡的影响。结果 DCA-Na能明显抑制A549细胞生长,呈浓度-时间依赖性;DCA-Na作用后电镜观察到典型的细胞凋亡形态学特征;Caspase 3试剂盒检测结果显示,DCA-Na可诱导A549细胞表达Caspase 3,其活性随DCA-Na剂量的增加而递增;与对照组比较,线粒体跨膜电位均明显下降,且去极化百分率随DCA-Na剂量及时间的增加而增加。结论 DCA-Na可诱导A549细胞株凋亡,线粒体跨膜电位下降可能是其诱导细胞凋亡的重要机制。Objective To research the function of small molecule drug-sodium dichloroacetate（DCA-Na） to the apopto- sis in vitro of antineoplastic and induced tumor cell in human lung adenocareinoma cell line-A549, and to explore the possible mechanism of anti-tumor effect. Methods Methyl thiazolyl tetrazolium （MTT） assay was adopted to observe the inhibition of A549 cell 24,48 and 72 h after treatment with different concentrations of DCA-Na. Flow eytometry,transmission electron micro scope （TEM） and Caspase 3 assay kit were used to observe the influence of DCA-Na to the apoptosis of A549 cell. Results DCA-Na could apparently inhibit the growth of A549 cell in a dose-time manner. After the conduction of DCA-Na,TEM could perceive the morphological characteristics of the apoptotic cell. Caspase 3 assay kit showed that DCA-Na could induce the expression of Cas- pase 3 in A549 cell, and activity degree of Caspase 3 increased with increase of the dosage of DCA-Na. Compared with the control group, mitoehondrial membrane potential in the treatment group decreased significantly, and the apoptosis rate increased in a dose-dependent manner by the contrast. Conclusion DCA-Na can induce the apoptosis in A549 cell,and the reduction of the MMP might play an important role in apoptosis.
