摘 要:将顺铂和磁性氧化铁共包于白蛋白中而制成了磁性靶向顺铂白蛋白微球。按正交设计筛选最佳合成工艺为:顺铂用量为40mg,白蛋白用量为250mg,FeCl2用量为282.8mg,pH=10。并以微球进行了质量考察:微球平均粒径为2.89±0.51μm,磁性靶向顺铂微球的载药量为13.64%(mg/mg),包裹率为85.24%;含药微球在体外12h释放完全,具一定的缓释作用;稳定性考察表明所制微球稳定性良好。Targeted and Magnetic cisplatnum albumin microspheres had been prepared by precipitation of magnetite from the mixture of ferrous ions and hydrogen peroxide in the presence of albumin and cisplatnum and sodium hydroxide. An optimum procedure was established by orthogonal test for preparing magnetic cisplatnum albumin microsphere: the mass concentration of albumin and ferrous chloride and cisplatnum were 250mg, 282. sing and 40mg respectively. pH value of solution was 10. The quality, stability, safety and drug loading were studied. The results showed that the mean size was 2. 89± 0. 51um, its embedding ratio was 85. 24%, its stability could be kept at 3-5℃ and room temperature for 3 months.
