摘  要:目的:探讨内皮素(ET)-1对原代培养的大鼠肾上腺皮质细胞增殖与分泌的影响,以期优化肾上腺皮质细胞体外培养方案,为肾上腺细胞移植提供最适宜的条件。方法:原代培养大鼠肾上腺皮质细胞,培养液中含不同浓度(10-6^-15mol/L)ET-1,以MTT法检测细胞增殖;细胞爬片免疫组化检测各组细胞增殖因子(PCNA)阳性率;放射免疫法检测各组培养液中皮质酮浓度。结果:ET-1浓度为10-10^-11 mol/L时,MTT法吸光度值、PCNA阳性率、皮质酮浓度均高于对照组(P<0.05)。结论:10-10^-11 mol/L ET-1可有效促进大鼠皮质细胞的增殖及分泌功能,增加细胞活力,利于肾上腺细胞移植的进行。[Abstract] Objective: To study the effect of endothelin(ET)-I on the proliferation and secretion of primary rat adrenocortical cells, and to optimize the culture condition and provide a better environment for adrenocortical cells transplantation. Methods: Primary rat adrenocortical cells were exposed to different concentrations (10-6-15 mol/L) of ET-1 in vitro. Adrenocortical cells proliferation was determined by MTT and immunocytochemistry with PCNA. Corticosterone concentrations in mediums were tested by radioimmunoassay. Results: Compared with the control, the adrenocortical cells exposed to 10-10-11 mol/L ET-1 exhibited significantly higher absorbency value, PCNA positive rate and cortieosterone concentration. Conclusion: In appropriate concentration(10-10-11mol/L), ET-1 can effectively boost proliferation and secretion of adrenocortical cells, and provide a better condition for adrenocortical cells transplantation.
