摘 要:高通量测序方法筛查媒介昆虫携带病原体是一种新的重要方法。研究表明小RNA高通量测序可用于昆虫携带病毒的检测。本工作尝试用小RNA高通量测序法筛查昆虫携带的病毒,发现该方法不仅可用于媒介昆虫携带病毒的检测,也可以用于原核和真核病原体的筛查。用该方法对野外采集的3种蚊虫和3种蜱进行分析,用blast程序对NCBI核酸序列库(nt)进行比对搜索,从测序结果中查找到多种病毒、原核及真核的病原体。上述结果表明,小RNA高通量测序可以用于各种病原体的发现,有望发展成为一种通用的病原体筛查技术。High-throughput sequencing technology has found applications in various research fields. The High-throughput sequencing of small RNAs has been effectively used in finding the viruses carried by vector insects. This work intended to use this technique on screening for vector-borne viruses, and prokaryotic and eukaryotic pathogens other than viruses were unexpectedly identified. The small RNAs from 3 wild-eaught mosquito specieses and 3 tick speeieses were sequenced, and the sequences were aligned and blasted against NCBI nueleotide database (nt). Several viruses, prokaryotic and eukaryotic pathogens were screened out from the blast results. Thus this strategy can identify possible pathogens carried by vectors and become a universal screening technique for survey various kinds of pathogens in insects.
