摘 要:目的:定性定量检测不同企业生产的紫草配方颗粒中的左旋紫草素,探讨配方颗粒的质量标准及与传统饮片的差异。方法采用薄层色谱法(TLC)定性鉴别紫草配方颗粒;用高效液相色谱(HPLC)测定紫草配方颗粒中左旋紫草素含量。结果 TLC鉴别中,紫外灯(365 nm)下检视,在与左旋紫草素对照品相应位置上,部分紫草配方颗粒显现相同的斑点;HPLC测定中,在0.106~1.272μg/mL范围内,左旋紫草素浓度与峰面积呈良好的线性关系,回归方程为y=13208x+65.067,r=0.9990;平均加样回收率为97.29%,RSD=3.20%(n=6)。不同厂家不同批次样品的左旋紫草素含量为0.09~0.23 g/g不等。结论紫草中药配方颗粒中左旋紫草素含量较低,不同厂家不同批次间的差异较大,需进一步研究以确定左旋紫草素含量与配方颗粒质量之间的关系。Objective To analyze qualitatively and quantitatively of L-shikonin in Lithospermum formula particles produced by different companies, and to discuss the difference between the quality standard of the formula particles and that of traditional Chinese medicine decoction pieces. Methods The ingredients in the Lithospermum formula particles were identified by thin-Layer chromatography (TLC). The content of L-shikonin in Lithospermum formula particles was determined by HPLC. Results As shown in TLC analysis, some Lithospermum formula particles showed spots at the position identical to that of L-shikonin reference substance observed under 365 nm UV Light. There is a good linear relationship between the concentration of L-shikonin and the peak area in the range of 0.106-1.272μg/mL (r=0.9990). The mean recovery is 96.55%, with RSD of 3.57%(n=6). Conclusion The content of L-Shikonin of Lithospermum formula particles is low. There is obvious difference among the levels of L-Shikonin of different batches of products or products provided by different manufacturers. Further research is needed to determine the relationship between the content of L-Shikonin and the quality of formula particles.
