摘  要:目的:通过观察针刺"百会"透"曲鬓"穴头针疗法对急性脑出血大鼠脑组织形态学及脑组织中β-catenin表达的影响,研究针刺对脑出血后NSCs增殖分化的影响及其与Wnt信号通路关键因子β-catenin的关系,阐明针刺促进脑出血后神经重塑的作用机制。方法:选用雄性Wistar大鼠按随机数字表法随机分为空白组、模型组、针刺组、通路激动剂(SB216763)组;每组再随机分为1天、3天、7天、14天4个亚组。模型组采用立体定向手术自体血注入法制备脑出血模型;针刺组大鼠针刺患侧"百会"透"曲鬓"穴。在光镜下观察各组大鼠脑组织形态学改变;运用蛋白免疫印迹方法检测和免疫组化方法检测各组大鼠脑组织β-catenin的阳性细胞表达以及针刺治疗对它们的影响。结果:针刺及激动剂组可明显改善大鼠神经功能缺损评分,与模型组比较,有显著性差异(P<0.05)。模型组、针刺组及SB216763组大鼠灶周组织β-catenin阳性细胞数表达均随时间点变化逐渐增加,3天时达高峰值,7天逐渐减少,14天时仍有表达;针刺组、SB216763组分别与模型组比较,差异均有统计学意义(P<0.05);针刺组与SB216763组中β-catenin表达比较无明显差异(P>0.05)。结论:针刺及通路激动剂均可明显改善脑出血大鼠神经损伤功能评分。"百会"透"曲鬓"针刺法可能具有与通路激活剂SB216763有相类似的作用,可能通过上调β-catenin表达,激活Wnt信号通路,参与调控脑出血大鼠脑组织中神经干细胞的增殖与分化。Objective:To observe the effect of acupuncture “Baihui” through “Qubin” head acupuncture effect on expression of β- catenin morphology of rats' brain tissue and brain tissue in the acute cerebral hemor- rhage, effect of acupuncture on the proliferation and differentiation of NSCs after intracerebral hemorrhage and its relationship with Wnt signal pathway key factor beta -catenin, elucidating acupuncture action mechanism to promote neural plasticity after cerebral hemorrhage. Methods: The rats were randomly divided into blank group, model group, acupuncture group, channel agonist male Wistar rats (SB216763) group; each group was randomly divided into 1 days, 3 days, 7 days, 14 days and four subgroups. The model group was by ster- eotactic operation of autologous blood preparation of cerebral hemorrhage model rats of acupuncture group acu- puncture injection ; side “Baihui” through “Qubin” point. Morphological observation of brain tissue of rats inthe changes under light microscope; using Western blotting and immunohistochemistry method to detect the effect of positive cell expression in brain tissue in rats of β - catenin and acupuncture treatment on them. Re- suits: Acupuncture and agonist group could significantly improve the neurological deficit scores of rats, com- pared with model group, there were significant differences (P 〈 0.05 ). Immunohistochemistry showed : model group, acupuncture group and SB216763 group β- catenin positive cells in peripheral tissues of rats with focal expression of time change gradually increased, reached the peak value at 3d, 7d decreased gradually, there when 14d expression; acupuncture group, SB216763 group and model group were compared, there was a sta- tistically significant difference ( P 〈 0.05 ) ; acupuncture group and SB216763 group 13 - catenin expression showed no significant difference (P 〉 O. 05 ).
