摘 要:目的建立一种临床粪便标本的前处理方法,使之与免疫层析技术相结合,用于沙门菌引起的食物中毒临床标本的快速检测。方法采用密度梯度离心、煮沸、免疫磁珠等技术相结合,处理人工污染的标本。方法一是采用样品经免疫磁颗粒富集菌体再经过60℃水浴30 min处理。方法二是经梯度离心方法处理再经煮沸30 min。然后使用沙门菌胶体金免疫层析试纸条进行检测,根据检测结果,确定最佳的前处理方法。结果两种方法相比检测结果无差别,检出限均为105CFU/ml。可从时间、和费用方面来说,经梯度离心方法处理再经煮沸30 min的方法操作更简便、实用。结论本实验建立了一套临床粪便标本的前处理方法,成功应用于沙门菌引起的食物中毒临床标本的快速检测。检测速度快,操作简便。Objective To establish a pretreatment method of clinical fecal specimens combining with immunochromatographic assay for rapid detection of Salmonella in food poisoning. Methods Density gradient centrifugation, boiling, immunomagnetic bead technology were combined to treat the artificial - contamination specimens. In accordance with the first method, the bacteria in samples was enriched by immunomagnetic particles, then treated in 60℃ waterbath for 30 rain. Another method was using gradient centrifugation treatment followed by 30 min boiling. Then salmonella in samples was detected by colloidal gold immunochromatographic strip. The optimal pretreatment method was selected according to the test results. Results The test results of the two preatment methods had no difference, and the detection limits were both 105 CFU/ml. But the gradient eentrifugation combining with 30 min boiling for pretreatment was more convenient and practical in view of time and cost. Conclusion A set of pretreatment method has been established in this study for rapid detection of clinical specimens from food poidoning. This method was easy to operate and rapid.
