摘  要:为研究华癸中慢生根瘤菌(Mesorhizobium huakuii)7653R外膜蛋白Opa22在共生固氮过程中的作用机制,构建了细菌双杂交基因组随机文库,并以Opa22为诱饵,钓取与其相互作用的候选靶蛋白。结果获得12个与Opa22蛋白互作的阳性克隆,为后续研究Opa22的作用机制提供了新的线索和思路,同时也为开展根瘤菌蛋白互作的研究构筑了良好的技术平台。Opa22 is an outer membrane protein identified in Mesorhizobium huakuii 7653R in our previous work. To investigate its functions during symbiotic nitrogen fixation, a random genomic DNA library of M. huakuii 7653R based on the bacterial two-hybrid system was constructed. The candidate proteins interacting with Opa22 were screened from the library and twelve positive clones were obtained and analyzed. This study will provide both new insights on understanding Opa22 functions during symbi- osis and technique platforms for the further researches on the protein-protein interaction network as well.
