摘  要:目的 探讨云芝多糖对人CIK细胞增殖及细胞杀伤功能相关分子表达的影响.方法 体外培养CIK细胞,不同浓度云芝多糖诱导人CIK细胞24h,CCK-8比色法检测人CIK细胞增殖,流式细胞仪测定CIK细胞表达的穿孔素、颗粒酶B和CD107a的变化.结果 不同浓度的云芝多糖可促进CIK细胞增殖,促进CIK细胞穿孔素、颗粒酶B的和CD107a的表达,25 mg/L云芝多糖作用最强;但随着药物浓度的增加,CIK细胞增殖率逐渐降低,且细胞杀伤功能相关分子表达含量下降.结论 云芝多糖在低浓度下可促进CIK细胞增殖,提高CIK细胞的杀伤活性和功能.Objective To explore the effects of coriolus versicolor polysaccharide (PSK) on the proliferation of human CIK cells and the expression of cytotoxicity associated molecules. Methods PSK at different concentrations was used to induce the growth of CIK cells in vitro for 24 hours. Cell proliferation was detected by the CCK8 assay. The changes of perforin, GraB and CD107a in CIK cells were examined by flow cytometry. Results PSK could stimulate the proliferation of CIK cells and increase the expression of perforin, granzyrne B, and CD107a in CIK cells. 25 μg/mL PSK produced the strongest effect (P 〈 0.05 ). With the rising of PSK concentrations, the growth rates of CIK cells were reduced while the content of cyotoxicity associated molecules was decreased. Conclusion Low concentrations of PSK can promote the growth of CIK cells and improve their cytotoxicity and function.
