摘 要:以南方泡桐( Paulownia australis)二倍体及其同源四倍体幼苗为材料,分别采用AFLP和MSAP分子标记技术研究其DNA碱基序列及其甲基化的变化情况。研究结果表明:AFLP中平均每对引物扩增出50~70条谱带,南方泡桐二倍体染色体加倍前后DNA碱基序列在AFLP水平上没有发生变化;统计MSAP在100~500 bp的扩增带,二倍体南方泡桐与四倍体南方泡桐酶切位点分别有2059和2214个,发生甲基化的位点分别为34.34%和36.77%,而发生DNA全甲基化的位点则分别为9.76%和11.74%。四倍体南方泡桐与二倍体南方泡桐相比40.80%位点甲基化模式发生了变化,并且同源四倍体南方泡桐的总甲基化水平和半甲基化水平比其二倍体高。By the amplified fragment length polymorphism ( AFLP ) and the methylation-sensitive amplification polymorphism ( MSAP) , the experiment was conducted to analyze the first structure of total DNA and the levels and patterns of methyla-tion between diploid and autotetraploid Paulownia australis seedling, respectively.Each pair primer can amplify 50-70 bands on average with AFLP, these amplification sites of diploid P.australis are identical to its autotetraploid .The MSAP can amplify fragment from 100 to 500 bp.The 2 059 and 2 214 sites are in diploid and autotetraploid P.australis, and the methylation sites are 34.34% and 36.77% of total methylation sites, respectively, where fully DNA methylation rate of them are 9.76% and 11.74%.By comparing autotetraploid P.australis with diploid, 40.80% sites show changes of DNA methylation paterns, and the global DNA methylation level and hemi-methylation level of autotetraploid are higher than those of diploid.
