摘 要:目的探讨白藜芦醇(Res)对大鼠心肌缺血-再灌注(I/R)后心律失常的影响及其与Ca2+-ATP酶(SERCA2)的关系。方法将SD大鼠随机分为假手术组、I/R后心律失常模型组、Res低、高剂量干预组(10、30 mg·kg-1,bid)。ig给予Res3 d后,制造I/R心律失常模型,用Power-Lab系统检测心电图的变化并进行心律失常评分,记录各组大鼠因心律失常导致的死亡数;用Real time PCR法检测心肌样本中肌浆网Ca2+-ATP酶中mRNA表达的变化。结果与模型组比较,Res组能显著减轻心室纤颤的发生率和持续时间,改善心律失常评分;上调SERCA2中mRNA的表达。结论 Res可明显减轻大鼠心肌I/R后心室纤颤,其机制至少可能与其减轻心肌细胞Ca2+超载相关。OBJECTIVE To observe the effect of resveratrol (Res) on reperfusion arrhythmia in the rats and to explore the mecha- nism. METHODS The SD rats were randomly divided into the sham - operated group and model group, the low and high dose of Res treatment group( 10 and 30 mg.kg^-1 ,ig,bid). After Res were administered for 3 d,the I/R arrhythmia model as made. ECG was recor- ded with the power - lab experiment system. The mRNA expression of SERCA2 in the rat' s myocardium was evaluated by real time PCR method. RESULTS Compared with that of the model group, the rate and durations of ventricular fibrillation were markedly reduced, and arrhythmias values were significantly improved in Res - treated group. Compared with that of the model group, the mRNA expression of SERCA2 could be markedly increased in Res - treated group. CONCLUSION Res can significantly alleviate the arrhythmia after myocardial ischemia - reperfusion. The mechanism may be at least associated with the effect of reducing intracellular Ca2 + overload.
