摘  要:目的探索建立1-氯-2,4-二硝基苯(DNCB)经皮肤致敏诱导BALB/c小鼠特应性皮炎(AD)模型的方法。方法将6只7周龄BALB/c小鼠随机分为3组(n=2),常规饲养。3组小鼠分别接受1%+0.2%DNCB溶液(A组)、1%+0.1%DNCB(B组)和基质(橄榄油:丙酮=4:1)溶液(正常对照C组)处理。用上述溶液反复刺激小鼠的背部和耳廓皮肤,观察皮炎症状和皮肤病理等指标,并检测血浆总IgE浓度。结果在首次致敏后的4周中,B组小鼠出现明显皮炎症状,A组小鼠出现轻微皮炎症状后恢复正常,C组无明显皮炎症状。B组小鼠的抓挠动作次数、皮炎症状评分显著高于A组和C组。皮肤病理观察显示B组棘层细胞增厚、炎症细胞浸润,其嗜酸性粒细胞、单核细胞、淋巴细胞、中性粒细胞和肥大细胞较其它两组有显著差异。B组血浆总IgE浓度高于A、C两组,但样本例数过少,尚未得到统计学支持。结论先后用含1%和0.1%DNCB的丙酮-橄榄油溶液涂拭BALB/c小鼠皮肤能够成功建立AD模型。Objectives To establish a BALB/c mice model of atopie dermatitis (AD) induced by 1-chloro-2,4-dinitrobenzene (DNCB). Methods Six BALB/c mice (7 weeks old) were divided into three groups (n = 2) with normal feeding. The three groups of mice were treated with 1% and 0.2% DNCB solution (group A),1% and 0.1% DNCB solution (group B),and matrix (1:4 mixture of acetone and olive oil) solution (group C for control) , respectively. These solutions were besmeared onto mice dorsal and ear skin repeatedly ,and the dermatitis symptoms, skin pathology, and total plasma IgE were evaluated. Results In the 4 weeks' induction period,the dermatitis symptoms were obviously presented in group B, and mildly occurred and got recovered in group A. No obvious symptoms were observed in the control group. The dermatitis severity score and scratch count of group B was significantly higher than those of group A and group C. As indicated by the skin pathology,stratum spinosm was thicker,inflammatory cell infiltration was more obverse,and the numbers of eosinophils,monocytes,lymphocytes,neutrophils,and mast cells were significantly higher in group B than in the other two groups. Due to the small number of sample size,the professionally higher total plasma IgE concentration in group B was not yet supported by statistics. Conclusions Repeated application of DNCB on the dorsal skin of BALB/c mice induced AD-like symptoms, which could be a useful animal model for atopic dermatitis research.
