摘 要:目的:研究丹参酚酸类化合物的体外抗氧化暨对帕金森病细胞模型的神经保护作用。方法:应用MTT比色法检测酚酸类化合物对过氧化氢以及1-甲基-4-苯基吡啶阳离子(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,MPP+)诱导的PC12细胞损伤模型的存活率影响;应用全自动生化分析仪检测活性最强的原儿茶酸(protocatechuic acid,PAc)对MPP+诱导的PC12细胞损伤模型的乳酸脱氢酶释放的影响;应用高内涵筛选系统检测PAc对MPP+诱导的PC12细胞损伤模型的凋亡多参数(F-actin和线粒体膜电位)的影响。结果:Pac 200 mg·L-1的抗氧化活性最强,它可抑制MPP+诱导的乳酸脱氢酶释放和凋亡多参数改变。结论:PAc具有抗PC12细胞氧化损伤作用。Objective: To determine the in vitro anti-oxidative and neuroprotective effect of phenolic acid compounds (PAc) from Danshen in a cell model of Parkinson's disease (PD). Methods: After exposure to 1- methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPP^+) , cell viability was detected by MTT assay. The release of lactate dehydrogenase (LDH) was inspected by an antomatic analyzer. The multiple parameters of apoptosis (nu- cleus, F-actin and mitochondrial membrane potential) were analyzed by high content screening (HCS). Results: Protocatechuic acid (Pac, 200 mg· L^-1) had the strongest in vitro anti-oxidative and neuroprotective effect in MPP^+-treated PC12 cells. PAc ameliorated the release of LDH and the changes in apoptosis parameters in the PC12 cells. Conclusion: PAc can inhibit the oxidative damage in PC12 cells.
