摘  要:目的:探讨祛瘀生肌中药对糖尿病难愈合性创面修复MAPK信号通路相关基因的调控作用。方法:以糖尿病溃疡创面为研究平台,将SD大鼠随机分为正常创面组、糖尿病溃疡模型组及生肌化瘀1(生肌方与化瘀方2∶1)、生肌化瘀2(生肌方与化瘀方1∶1)、生肌化瘀3(生肌方与化瘀方1∶2)方组,除正常组外,其余4组以腹腔注射链脲佐菌素(STZ)法制备糖尿病模型。每组给予相应干预措施后,采用Real-Time PCR技术检测MAPK信号通路应激活化蛋白激酶(JNK)、活化蛋白激酶磷酸酶(MKP)以及磷酸化p38、p53、p21 mRNA表达及分布规律。结果:与糖尿病溃疡模型组比较,祛瘀生肌中药组均可下调成模第7、11、15天MKP、JNK mRNA表达,上调p53、p38 mRNA表达,p21 mRNA表达没有明显改变,且各组间无明显的作用规律。结论:MAPK信号通路在促进糖尿病溃疡创面愈合中发挥重要的作用,祛瘀生肌中药可调控MAKP通路相关基因。Objective:To explore the regulatory effect of Chinese medicine for stasis-removing and granulation-regeneration to repair the related gene regulation in MAPK signaling pathways in the refractory wounds of diabetes.Methods:Diabetic ulcer wounds were used as the research platform,72 SD rats were randomly divided into normal wound group,diabetic ulcer model group,formula 1 group( the ratio of prescription in stasis removing and granulation regeneration is 2:1),formula 2 group( the ratio is 1:1),and formula 3 group( the ratio is 1:2).Except the normal wound group,the other groups were treated with streptozotocin( STZ) by intraperitoneal injection to duplicate the diabetic models.After giving corresponding intervention measures,we detected the expression and distribution of JNK,MKP,phosphorylated p38,p53 and p21 mRNA in the MAPK signaling pathway by Real-Time PCR.Results:Compared with diabetic ulcer group,the expressions of MKP and JNK mRNA were down-regulate and expressions of p53 and p38 mRNA were up-regulated in formula groups.But no significant difference was found in expression of p21 mRNA.No remarkable difference was found among formula groups.Conclusion:MAPK signaling pathway pays an important role in the wound healing of diabetic ulcers,and the prescription of stasis removing and granulation regeneration can regulate the related genes in MAPK signaling pathway.
