摘  要:目的建立雪貂感染H7N9禽流感病毒动物模型。方法A/Anhui/1/2013(H7N9)禽流感病毒经鼻吸入感染雪貂,观察动物临床症状和体征,上呼吸道排毒情况及组织病理学变化。结果雪貂感染后出现体重下降、活动减少以及打喷嚏的临床表现,上呼吸道、心脏、肝脏及嗅球可检测到活病毒,上呼吸道排毒的峰值出现在感染后的第3—5天。血清抗体滴度最高达到1280,外周血淋巴细胞数量减少、粒细胞数量增加。组织病理学显示动物肺脏呈局灶性间质性肺炎及肺泡炎改变,CT显示肺内片状阴影。结论成功建立雪貂感染H7N9禽流感病毒的动物模型,模型的建立为H7N9禽流感发病机制研究、药物及疫苗的评价奠定了实验基础。Objective To establish ferret models for the novel avian-origin H7N9 influenza virus. Methods A/ Anhui/1/2013 (H7N9) virus was administered by intranasal instillation to groups of ferrets. Clinical signs, virus shedding from respiratory tract and pathological analyses were recorded. Results Inoculated ferrets developed typical clinical signs including body weight loss and sneezing. Peak virus shedding from respiratory tract was observed on 3 - 5 days post inoculation (d. p. i. ). Virus could also be detected in heart, liver, and olfactory bulb from inoculated ferrets. The inoculation of H7N9 could elicit seroconversion titers up to 1280. Leukopenia, significantly reduced lymphocytes but increased neutrophils were also observed in ferret models. Conclusion Collectively, the ferrets models enables detailed studies of the pathogenesis of this illness and lay foundation for relative drug or vaccine evaluation.
