摘 要:目的 :观察研究以药食两用中药红花、茯苓、甘草、大枣组成的复方红苓肝宝 ,对 CCl4造成小鼠慢性肝损伤肝纤维化形成的预防作用。方法 :40 % CCl4花生油溶液按 0 .1ml/10 g皮下注射 ,连续 5周 ,造成小鼠慢性肝损伤致肝纤维化病理模型 ,同时另设红苓肝宝和大黄虫丸防治组进行观察 ,检测肝重指数 ,转氨酶 (AST和 A LT) ,血清蛋白 ,肝羟脯氨酸 ,肝组织病理学改变和胶原纤维的相对量。结果 :单纯用 CCl4作模型组的小鼠 ,5周后与对照组相比 ,肝重指数明显增加 ,血清 A LT、AST的活性显著增强 ,肝组织羟脯氨酸的含量增高。病理组织学检查可见肝细胞出现大量变性、坏死 ,胶原纤维增生程度显著增加 ,出现了典型的慢性肝损伤后肝硬化的早期表明 ;应用红苓肝宝后 ,可使中毒小鼠转氨酶下降 ,白蛋白合成回升 ,球蛋白含量下降 ,A/G比值恢复 ,还可使肝脏中的羟脯氨酸含量显著下降 ,病理证明可使中毒小鼠肝脏细胞的变性坏死明显减轻 ,汇管区和肝小叶内纤维组织增生明显减少。结论 :红苓肝宝有降酶和护肝作用 ,可作为防止慢性肝炎所致的肝纤维化的保健食品应用。Objective:To determine the preventive effect of Hongling Ganbao (HLGB) on hepatic fibrosis induced by carbon tetrachloride (CCl 4) in mice. The HLGB consists of Chinese material medicine: safflower、tuckahoe、liquorice、jujube etc. which can be used for healthy food and medicine. Methods: Forty PCR mice were randomizedly divided into 4 groups with 10 mice in per group. First group was as normal control, the other three groups were given sc injection of 40% CCl 4 every 5 days for 5 weeks to induce chronic liver damage and fibrosis. Then they were either orally treated with saline as pathological model group, 2.2 g/kg HLGB or 0.78 g/kg Dahuang Zhechong Pill (Dhzcw, country medicine as a positive control) for 5 weeks, the later two groups were treated in the beginning day of CCl 4 administration. At the end of the study, mice were killed and the liver tissue was prepared for morphology to observe the degrees of inflammatory activity、fibrous hyperplasia、collagenous fiber relative capacity、hydroxyproline (HyP) and the liver weight index number (g/10 g); also to observe the levels of serum alanine aminotransferase (ALT)、 aspartic acid aminotransferase (AST)、total protein (TP)、albumin (Alb) and immunoglobulin (Ig). Results: serum ALT、AST、Ig and HyP of liver tissue were significantly increased in the model group compared to the normal group. The morphology sections showed that most of the hepatic cells in model group were serious degeneration, the typical cirrhosis occurred after chronic hepatitis injury. When compared to the model group, HLGB can markedly reduce the activity of the serum ALT and AST, decreased Ig and HyP, increased the serum Alb and the A/G. The morphology observation proved that the HLGB group was much better than the DHZCP treated group. Conclusion: HLGB has both the effects of reducing the serum aminotransferase and protect the hepatic tissue from CCl 4 damaged.
