摘 要:目的测定绝经后妇女血清基质金属蛋白酶-10(MMP-10)和金属蛋白酶抑制物-1(TIMP-1)水平,并探讨其与骨密度数值和骨代谢指标的关系。方法选取武汉地区120名48岁至65岁绝经后女性,用酶联免疫吸附试验(ELISA)测定血清MMP-10、TIMP-1以及整合素(Integrin)α2β1、雌二醇(E2)、骨保护蛋白(OPG)、骨保护蛋白配体(OPGL)的水平,计算MMP-10/TIMP-1比值,用双能X线吸收法(DEXA)测定腰椎正位,股骨颈,华氏区和大粗隆的骨密度(BMD)。同时,按照WHO标准将入选女性分为骨密度正常组、低骨量组和骨质疏松组。结果①骨质疏松组中血清MMP-10检测结果(644.25±19.21)pg/ml高于正常组(347.08±21.41)pg/ml(P<0.05),而TIMP-1检测结果(134±116)μg/L低于正常组(146±130)μg/L。②骨质疏松组中血清MMP-10与骨密度、血清E2、OPGL和Integrinα2β1测定结果存在明显负相关性(P<0.05),和OPG和CTX存在明显正相关性(P<0.05)。结论血清MMP-10和MMP-10/TIMP-1比值与绝经后骨质疏松症妇女骨密度和骨代谢指标具有关联性。两者可能通过整合素途径参与绝经后骨质疏松症骨代谢高转换过程。Objective To determine the serum levels of matrix metalloproteinase-10 ( MMP-10 ) and tissue inhibitor of matrix metalloproteinase-1 ( TIMP-1) in postmenopausal women, and to explore the correlation of the serum levels of MMP-13 and TIMP-1 with the bone mineral density ( BMD) and bone metabolic markers.Methods One hundred and twenty postmenopausal women in Wuhan area, aging from 48 to 65 years old, were selected.The serum levels of MMP-10, TIMP-1, integrinα2β1, E2, OPG, and OPGL of all the subjects were detected using ELISA.And the ratio of MMP-10/TIMP-1was calculated.BMD of the lumbar vertebrae, the femoral neck, the Ward ’ s area, and the greater trochanter was detected using dual energy X-ray absorptiometry ( DEXA) .Then, all the selected subjects were divided into 3 groups according to the WHO criteria:the normal control group, the low bone mass group, and the osteoporosis group.Results The serum concentration of MMP-10 in osteoporosis group (644.25 &#177; 19.21 pg/ml) was significantly higher than that in normal control group ( 347.08 &#177;21.41 pg/ml, P&lt;0.05 ) , while the serum concentration of TIMP-1 was lower in osteoporosis group (134 &#177;116 μg/L) than that in normal control group (146 &#177;130 μg/L). In osteoporosis group, the notable negative correlations between MMP-10, and BMD, serum E2 , OPGL, and integrin α2β1 were observed (P&lt;0.05).Positive relations were observed between MMP-10, OPG, and CTX (P&lt;0.05).Conclusion The serum level of MMP-10 and the ratio of MMP-10/TIMP-1 are significantly correlated with BMD and bone metabolic markers in postmenopausal osteoporosis women.They may participate in the high-speed turnover of bone metabolism in postmenopausal&amp;nbsp;osteoporosis women through integrin signal pathway.
