摘  要:目的探索miR-92a介导的耳蜗毛细胞放射性损伤机制。方法运用缩小RNA(miRNA)芯片技术检测耳蜗毛细胞株HEI-OC1放射前后miRNA的表达谱差异,qRT-PCR验证miR-92a在细胞照射后表达变化情况,对细胞转染阴性对照物、miR-92a模拟物及抑制物后,用MTT法检测耳蜗毛细胞放射后增殖能力的变化。结果芯片及qRT-PCR验证结果显示耳蜗毛细胞照射后miR-92a表达量明显升高(P<0.05);与阴性对照相比,转染了miR-92a模拟物的细胞在照射(2、4、8、16 Gy)后细胞活性明显下降(P<0.05),转染了miR-92a抑制物的细胞在照射(8、16 Gy)后细胞活性明显上升(P<0.05)。结论 miR-92a的过度表达加重了毛细胞的放射损伤,miR-92a可能是耳蜗毛细胞放射性损伤重要的调节因子。Objective To investigate the mechanisms of radiation - induced cochlea hair cell injury mediated by miR - 92a. Methods Microarray and qRT - PCR were performed for assessment of the expression of microRNAs in the cochlea hair cell line HEI - OC1 between before and after radiation. MTT assay was performed on cochlea hair cells trans- feeted with negative control, miR -92a mimics or inhibitor post -radiation. Results The expression of miR -92a was significantly up - regulated after radiation ( P 〈 0. 05 ). Compared with ceils transfected with negative control, significantly lower and higher post - radiation viabilities were observe in cells transfected with miR - 92a mimics and miR - 92a inhibitor, respectively ( P 〈 0. 05 ). Conclusion The over - expression of miR - 92a intensifies cochlea hair cell injury, which may be an important regulator in radiation -induced cochlea hair cell injury.
