摘  要:目的探讨不同锰累积暴露量的工人DNA损伤修复基因XRCC1 C26304T位点多态性与神经行为改变的关系。方法选取200名半年以上工龄的职业性锰暴露冶炼工人和94名劳动强度相似但无锰暴露的工人,按照累积暴露指数将观察对象分为高、低暴露组和对照组。利用垂直沟槽稳定试验和插板试验进行神经行为改变的检测;提取血白细胞DNA,利用PCR-RFLP法检测XRCC1 C26304T位点的单核苷酸多态性(SNPs)。结果高、低暴露组携带XRCC1 C26304T位点变异基因型TT的工人在垂直沟槽稳定试验的划痕长度、插板试验的得分上均低于携带野生基因型CC和杂合基因型CT的工人,碰撞次数高于野生基因型和杂合基因型(P<0.05);对照组内各基因型之间无统计学差异(P>0.05)。结论 XRCC1C26304T位点的多态性可能影响职业锰暴露者的神经行为;XRCC1 C26304T位点的多态性有可能作为锰致神经行为改变以及锰中毒风险评估的检测指标。Objective To study the relationship between neurobehavioral changes and XRCC1 gene polymor- phism in manganese exposured workers. Methods 200 Mn exposured workers with working age 〉 0.5 year and 94 Mn non- exposured workers were chosen and divided into high and low exposure groups; and control group ac- cording to cumulated exposure index (CEI). Every worker was examined by neurobehavioral detective equip- ment. The genetic DNA was extracted, and the genotypes of exon 6 polymorphism were detected by PCR - RFLP. Results In high and low exposured groups, there was a significant difference in subjects with different genotype of C26304T in three neurobehavioral tests (P 〈 0.05 ). In high and low exposured groups, the subjects with Tr genotype were significantly lower in the length of drawing and the scores of pegboard test than those with CC and CT genotypes ( P 〈 0.05 ), while collision frequencies were significantly higher than those with CC and CT genotypes (P 〈 0.05 ). In the control group, there was no significant difference in subjects with different genotypes in three neurobehavioral tests (P 〉 0.05 ). Conclusion XRCC1 C26304T polymorphismchanges in manganese -exposed workers; XRCC1 be used as an indicator to detect the manganese - induced neurobehavioral changes, and in manganese poslonlng. C26304T SNPs might as the risk assessment
