摘  要:目的:探讨α-硫辛酸(α-LA)对ApoE基因敲除小鼠血清明胶酶[基质金属蛋白酶(MMP)-2、MMP-9]的影响。方法:30只ApoE基因敲除小鼠按数字法被随机均分为高脂模型组、α-LA组、辛伐他汀组,于高脂饮食16周后,分别给予生理盐水、α-LA、辛伐他汀灌胃,另取10只G57BL/6J小鼠,于正常饮食后给予生理盐水灌胃作为正常对照组。8周后测血清MMP-2、MMP-9值。结果:与高脂模型组比较,α-LA组小鼠血清MMP-2[(680.24±49.25)μg/L比(528.47±56.64)μg/L]、MMP-9[(745.84±56.43)μg/L比(530.77±64.25)μg/L]水平下降,差别有显著性(P均<0.05),但与辛伐他汀组比较无显著差异(P>0.05)。结论:α-硫辛酸与辛伐他汀相似,可抑制血清明胶酶水平,起到稳定动脉粥样硬化斑块的作用。Objective:To explore influence of α-lipoic acid (α-LA) on serum gelatinases contents [matrix metalloproteinase (MMP)-2,MMP-9] in ApoE gene knockout mice.Methods:A total of 30 ApoE gene knockout mice were randomly and equally divided into high fat model group,α-LA group and simvastatin group according to number table.After high fat diet for 16 weeks,the three groups received normal saline,α-LA and simvastatin via intragastric administration respectively.Another 10 G57BL/6J mice,who received normal saline via intragastric ad ministration after normal diet,were enrolled as normal control group.Serum levels of MMP-2 and MMP-9 were measured after eight weeks.Results::Compare with high fat model group,there were significant decrease in serum levels of MMP-2 [(680.24 ± 49.25) μg/L vs.(528.47 ± 56.64) μg/L] and MMP-9 [(745.84 ± 56.43) μg/L vs.(530.77 ± 64.25) μg/L] in α-LA group (P〈0.05 both),but without significant difference between α-LA group and simvastatin group.Conclusion:The α-lipoic acid could decrease serum gelatinases levels and possess the effect stabilizing atherosclerotic plaques.
