摘 要:目的观察维生素E对高脂血症小鼠辅助性T淋巴细胞1(Th1)和辅助性T淋巴细胞2(Th2)细胞因子表达的影响。方法将50只ICR雄性性成熟小鼠随机分为A、B、C、D、E 5组,每组10只,分别给予高脂饲料、不同浓度维生素E(每日10 mg/kg、20 mg/kg、40 mg/kg)加高脂饲料及普通饲料连续喂养4周,后处死小鼠抽取静脉血观察比较血清Th1和Th2细胞因子表达情况[肿瘤坏死因子-α(TNF-α)、白细胞介素-12(IL-12)及白细胞介素-4(IL-4)、白细胞介素-5(IL-5)]及甘油三酯(TG)、胆固醇(TC)浓度。结果 A组血清TNF-α、IL-12及TG、TC浓度均高于B、C、D、E组,血清IL-4及IL-5浓度均低于B、C、D、E组,差异均有统计学意义(P<0.05)。B、C、D 3组血清TNF-α、IL-12及TG、TC浓度均随维生素E剂量增加逐渐降低,血清IL-4及IL-5浓度随维生素E剂量增加逐渐升高,且3组间比较差异均有统计学意义(P<0.05)。E组血清TNF-α、IL-12及TG浓度均低于B、C、D组,血清TC浓度低于B、C组,血清IL-4及IL-5浓度均高于B、C、D组,除D、E 2组血清IL-4比较差异无统计学意义外(P>0.05),余差异均有统计学意义(P<0.05)。血清IL-5浓度与IL-12浓度呈负相关关系。结论维生素E能抑制高脂血症所导致的免疫损伤与炎性反应,并可降低血脂。Objective To explore the effect of Vitamin E on expressions of helper T cells (Thl and Th2) in mice with hyperlipidemia. Methods A total of 50 viripotent male ICR mice were randomly divided into group A, B, C, D and E (n = 10). Mice fed on high-fat food in group A, B, C and D, and group B, C and D were lavaged with Vitamin E in different concentrations (10 mg/kg, 20 mg/kg and 40 mg/kg at each day) respectively for 4 weeks, while control group was fed on normal diet for 4 weeks, and then all the mice were sacrificed. The expressions of Thl and Th2 cytokines [ tumor necrosis fac- tor a (TNF-α) , interleukin 12 (IL-12) , interleukin 4 (IL-4) and interleukin 5 (IL-5)] and the concentrations of triglyeer- ide (TG) and cholesterol (TC) in serums of venous blood in the five groups were detected and compared. ReSults The con- centrations of TNF-α, IL-12, TG and TC in group A were significantly higher than those of group B, C, D and E (P 〈 0.05 ) , while the eoncentrations of IL-4 and IL-5 in group A were significantly lower than those of group B, C, D and E ( P 〈 0.05 ). In group B, C and D, the concentrations of TNF-α, IL-12, TG and TC were decreased gradually with increasing con- centration of Vitamin E, while the concentrations of IL-4 and IL-5 were increased gradually with increasing concentration of Vitamin E, and the differenees among the three groups were statistically significant (P 〈 0.05 ). In group E, concentrations of TNF-et, IL-12, and TG were lower than those of group B, C and D, TC concentration was lower than those of group B and C, concentrations of IL-4 and IL-5 were higher than those in group B, C and D, and the differences were statistically significant ( P 〈 0.05 ) except for the difference of serum IL--4 concentration between group D and E ( P 〉 0.05 ).
