丹蛭降糖胶囊对糖尿病模型大鼠VEGF及血小板参数的影响

摘　　要：[目的]探讨丹蛭降糖胶囊对糖尿病模型大鼠血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)及血小板参数的影响。[方法]取大鼠58只,分为5组,除正常组外,其余组给予高脂饲料四周,按35mg·kg-1给大鼠腹腔注射链脲佐菌素(Strep to zoe in,STZ)溶液1次,造模成功后除模型组和正常对照组外,各组连续灌胃药物4周,取血测定血糖血脂,VEGF及血小板参数。[结果]大鼠造模后VEGF的表达明显升高,与正常对照组比较有显著性差异(P<0.01),给予丹蛭降糖灌胃后,VEGF的高表达明显降低(P<0.01),疗效优于吡格列酮组。造模后,模型组大鼠血小板计数(PLT)降低,血小板平均容积(MPV)、血小板分布宽度(PPW)增高,予丹蛭降糖方后PLT水平上调,MPV、PDW降低,丹蛭高剂量组与模型组比较具有统计学意义(P<0.05或P<0.01);血糖及三酰甘油水平明显下降,与模型组比较(P<0.01)。[结论]具有益气养阴活血作用的丹蛭降糖胶囊,可控制VEGF的高表达,调节血小板参数,降低血糖及三酰甘油水平。[Objective]The aim of this study was to investigate the effects of DJC（Danzhi Jiangtang Capsule） on VEGF and blood platelet in type 2 diabetes rodent model. [Methods]58 Wistar rats were randomly assigned to 5 groups. Al animals in the experimental group were fed with a high calorie diet for 4 weeks and were injected with streptozocin（STZ,35 mg·kg-1）, except the control group. When diabetes rodent model was successful y made, the animals in the experimental group were kept with intragastric administration for 4 weeks. After that, blood glucose, VEGF and blood platelet levels were measured for al animals.[Results]The expression of VEGF was significantly greater compared with the control group（ P〈0.01）. However, the expression of VEGF was significantly less in the animals treated with DJC（P〈0.01）, and even less than the group with Pioglitazone. The excretion of blood platelet decreased and the excretion of MPV and PPW was more before treatment. However, the excretion of blood platelet significantly was greater and the excretion of MPV and PPW were lower in the high dose,compared with the model group（ P〈0.05 or P〈0.01）.The levels of blood glucose and TG were also significantly less, compared with the model group（P〈0.01 or P〈0.05）.[Conclusions]These results indicate that DJC controls the over-expression of VEGF, regulates the ex-cretion of blood platelet, besides decreasing the level of blood glucose and TG effectively.
