摘 要:目的:观察益肾活血方对单侧输尿管结扎(UUO)大鼠中低氧诱导因子(HIF-1α)、血小板源性生长因子(PDGF-BB)、细胞间黏附因子1(ICAM-1)以及血管细胞黏附分子1(VCAM-1)表达的影响。方法:实验采用UUO大鼠造成肾纤维化动物模型,并以益肾活血方、氯沙坦灌胃治疗,分别在术后第7天、第14天、第21天采集肾脏标本,运用免疫组化法检测HIF-1α、PDGF-BB、ICAM-1、VCAM-1在肾脏中的表达。结果:假手术组大鼠各时间点内,肾组织HIF-1α、PDGF-BB、ICAM-1、VCAM-1蛋白表达均在正常范围内;模型组大鼠随时间延长,肾组织HIF-1α、PDGF-BB、VCAM-1蛋白表达逐渐增加,给药组大鼠肾组织HIF-1α、PDGF-BB、ICAM-1、VCAM-1蛋白的表达有所降低(P<0.01或P<0.05),益肾活血方组降低优于氯沙坦组。结论:益肾活血方能明显改善肾纤维化过程中的内源性低氧状态、减轻大鼠肾间质的炎症反应,从而达到延缓肾间质纤维进展的作用。Objective:To investigate the effects of Yishen Huoxue formula on the expression of hypoxia-in-duced factor-1α (HIF- 1α ), platelet-derived growth factor-BB (PDGF-BB), intercellular adhesion molecule (I-CAM-1) and vascular cell adhesion molecule-l(VCAM-1) in kidney of UUO rats. Methods:Renal fibrosis rats were constructed by unilateral urethral obstruction(UUO) and were treated with oral Tongluo Huoxue formula or losartan. Then kidney specimen were collected at 7,14 and 21 days after surgery,and expres-sion of HIF-lot,PDGF-BB,ICAM and VCAM-1 in kidney were determined by immunohistochemistry. Re-sults:At each of the specified time points,the expression of HIF-1α,PDGF-BB,ICAM and VCAM-1 of the sham-operation group were all in physiological ranges. The model group,expression of HIF-1α,PDGF-BB,I-CAM and VCAM-1 gradually increased during the specified period. The treated groups showed decreased expression of HIF-1α,PDGF-BB,ICAM and VCAM-I(P〈0.01 or P〈0.05),and the decrease was larger in the Tongluo Huoxue formula group than losartan group. Conclusion:Yishen Huoxue formula can significantly al-leviate the endogenous hypoxia state during renal fibrosis,reduce renal interstitial inflammation,and thus de-lay progression of renal interstitial fibrosis.
